摘    要:目的:研究shRNA干扰midkine对胃肿瘤细胞SGC7901生物学特性的影响。方法:构建midkine基因的特异性小RNA干扰质粒,采用脂质体2000转染SGC7901细胞,检测细胞的增殖和克隆形成能力以及细胞凋亡情况。结果:与对照组相比,转染重质粒后1—5天,SGC7901细胞增殖明显受到抑制(P〈0.01),细胞形成克隆数明显降低(P〈0.01),并且有明显的亚二倍体峰出现(P〈0.05)。结论:针对midkine基因的特异性小RNA干扰质粒在体外能有效抑制人胃癌细胞SGC7901 midkine表达,细胞增殖及克隆形成能力减弱,细胞凋亡增加,为midkine基因靶向治疗提供一定的实验依据。

The effect of midkine shRNA on proliferation and apoptosis of the gastric carcinoma cell SGC7901

Abstract:Objective : To determine the effect of knockdown of midkine expression by shRNA on chatacteristic of human gastric carcinoma cell SGC7901. Methods:Specific shRNA plasmids to midkine were constructed,then transfected into SGC7901 by lipofectamin 2000. The cell proliferation, clony formation and apoptosis were detected. Results: Compared with control, SGC7901 cells transfected the recombinant plasmid expressing midkine shRNA, froml d to 5d cell proliferation was effectively inhibited (P 〈 0.01 ), cell clones decreased and cell apoptosis was significantly promoted ( P 〈 0.05 ). Conclusion: shRNA targeting MK gene can inhibit gastric cancer cells growth and induce apoptosis. MK shRNA may be a promising novel and potential therapeutic strategy for gastric cancer.
