EGF和HGF体外诱导肝硬化患者骨髓间充质干细胞向肝细胞分化的实验研究

目的探讨表皮细胞生长因子（epidermal growth factor，EGF）和肝细胞生长因子（hepatocyte growth factor，HGF）诱导肝硬化患者骨髓间充质干细胞（MSCs）向肝细胞分化的可行性。方法取肝硬化志愿者骨髓，密度梯度离心法联合贴壁筛选法获取MSCs，分别用含有HGF、EGF、HGF＋EGF或不加生长因子的培养基进行培养。用流式细胞仪、免疫细胞化学法鉴定细胞表型，酶联免疫吸附法检测培养上清液中Alb水平。结果肝硬化患者骨髓MSCs生长良好，表型为CD29阳性，CD34阴性。诱导组细胞可在21～28d时，形态变为三角形、多角型或类圆形。诱导组细胞7d检测到AFP的表达，21、28d检测出CK18阳性，并以时间依赖方式产生Alb。未诱导组在以上时间点未检测到上述指标。结论HGF、EGF均能诱导骨髓MSCs分化为肝样细胞，两者在一定程度上具有联合作用。
