糖尿病大鼠心肌缺血再灌注血管紧张素变化

摘    要:目的探讨菊苣多糖对链脲菌素糖尿病大鼠心肌缺血再灌注血管紧张素影响。方法应用链脲菌素复制大鼠糖尿病模型,12周后,腹腔注射戊巴比妥钠麻醉下气管插管安装呼吸机,开胸结扎冠状动脉左室降支60 min后松开结扎线再灌注60 min;标Ⅱ导联全程记录心电图,并于再灌注60 min取血、心脏;检测肌酸激酶(CK)、乳酸脱氢酶(LDH)、血管紧张素Ⅰ(AngⅠ)、血管紧张素Ⅱ(AngⅡ)。结果糖尿病模型对照组血清CK、LDH、AngⅠ、AngⅡ值分别为5.98 U/mL、10.03 U/mL、790.4 ng/L、1 773.7 ng/L,而菊苣多糖干预组血清CK、LDH、AngⅠ、AngⅡ值分别为1.39 U/mL、7.35 U/mL、443.9 ng/L、 1 552.2 ng/L,均低于糖尿病模型对照组,差异均有统计学意义(P<0.01)。结论菊苣多糖通过抑制AngⅠ、AngⅡ的产生对链脲菌素糖尿病大鼠心肌缺血再灌注损伤具有保护作用。

Abstract:Objective To analyze the effect of Herba Cichorii glycocalyx on angiotensin in streptozotocin-induced diabetic rats with myocardial ischemic/reperfusion injury.Methods The diabetic model of rats was established by intraperitioneal injection of streptozotocin.Twelve weeks later,trachea cannula and installation of life-support machine were performed under the condition of anaesthesia with intraperitioneal injection of pentobarbitol sodium sixty minutes after the ligation,the left anterior desending coronary artery was loosened for a sixty minutes reperfusion.The changes of electrocardiogram in Ⅱlead during whole course of experiment were recorded.Sixty minutes after the reperfusion,blood samples of the rats were collected for detections of creatine kinase(CK),lactic dehydrogenase(LDH),angiotensin I(Ang I),and angiotensin Ⅱ(Ang Ⅱ).Results The level of CK,LDH,AngⅠ,and AngⅡ in the diabetic rats were 5.98 U/ml,10.03 U/ml,790.4 ng/L,and 1 773.7 ng/L,while the level of CK,LDH,AngⅠ,and AngⅡ in the rats with chicory polysaccharide intervention were 1.39 U/ml,7.35 U/ml,443.9 ng/L,and 1 552.2 ng/L,respectively,and significantly lower than those in diabetes model(P<0.01 for all).Conclusion The chicory polysaccharide has protective effect on myocardial injury induced by ischemic/reperfusion in streptozotocin-induced diabetic rats through decreasing production of Ang Ⅰ and Ang Ⅱ.
