摘    要:目的:探索EP受体拮抗剂AH6809对炎症导致的BV-2 小胶质细胞凋亡的影响。方法:将脂多糖加入体外培养BV-2 细胞中制备炎症模型,通过倒置显微镜观察不同程度EP受体拮抗剂AH6809干预BV-2 细胞的形态学变化,应用免疫荧光染色、流式细胞术、TUNEL染色与免疫印迹检测BV-2 细胞后凋亡的情况。结果:AH6809浓度<80 μmol/L 时,BV-2 细胞炎症反应减轻,无凋亡产生;AH6809浓度≥ 80 μmol/L 时,BV-2 细胞出现凋亡;AH6809浓度达160 μmol/L 时,BV-2 细胞凋亡数显著增多。结论:在脂多糖诱导BV-2 细胞的炎症实验中,AH6809抗炎症反应的安全浓度控制在80 μmol/L 范围内。

Effect of EP receptor antagonist AH6809 on BV-2 microglialapoptosis induced by inflammation*
Abstract:Objective To explore the effects of EP receptor antagonist AH6809 on the apoptosis of BV-2 microglial cellscaused by inflammation. Methods Lipopolysaccharide was added to the BV-2 cells in vitro to establish inflammation modeland the morphological changes of BV-2 cells in different concentration of AH6809 intervention were observed by invertedmicroscope. Immunofluorescence staining, flow cytometry, TUNEL staining and Western blotting were used to detect theapoptosis of BV-2 cells. Results When the concentration of AH6809 was <80 μmol/L, the inflammatory response of BV-2cells was alleviated but no apoptosis occurred. When the concentration of AH6809 was≥ 80 μmol/L, BV-2 cells began toundergo apoptosis ; when the concentration of AH6809 reached 160 μmol/L, the number of apoptotic BV-2 cells increasedsignificantly and reached a peak. Conclusion In the lipopolysaccharide-induced inflammation experiment of BV-2 cells, thesafe concentration of AH6809 for anti-inflammatory reaction is in the range of 80 μmol/L.
