miR-124在大鼠坐骨神经损伤后的表达及对雪旺细胞增殖与迁移的影响

目的探讨miR-124在大鼠坐骨神经损伤后的表达及对雪旺细胞增殖与迁移的影响。方法建立大鼠坐骨神经损伤模型,实时PCR方法检测坐骨神经损伤后0 d、1 d、3 d、7 d神经近侧断端miR-124的表达水平。设计合成miR-124模拟物,转染雪旺细胞;MTT实验检测过表达miR-124对雪旺细胞增殖的影响;Transwell实验检测过表达miR-124对雪旺细胞迁移能力的影响。结果坐骨神经损伤后,与0d相比,miR-124在1 d、3 d、7 d的表达水平均明显增加(P<0.01)。转染miR-124模拟物能够显著上调雪旺细胞miR-124的表达水平,促进雪旺细胞增殖与迁移(P<0.01)。结论miR-124在坐骨神经损伤后表达水平逐渐升高,可能参与坐骨神经损伤后神经再生过程。

Expression of miR-124 of rat injured sciatic nerve and its effect on proliferation and migration of Schwann cells

Objective To investigate the expression of miR-124 of rats after sciatic nerve injury and its effect on the proliferation and migration of Schwann cells.Methods The rat model of injury of sciatic nerve were established.The level of miR-124 at the proximal end of the injured sciatic nerve in rats was detected by real time PCR on 0 d,1 d,3 d and 7 d.MiR-124 mimics was designed and synthesized to transfect Schwann cells.MTT assay was used to detect the effect of overexpression of miR-124 on proliferation of Schwann cells.Transwell assay was used to detect the effect of overexpression of miR-124 on migration of Schwann cells.Results CAfter sciatic nerve injury,the expression level of miR-124 on day 1,3 and 7 was significantly higher than that on day 0(P<0.01).After miR-124 mimic transfection,the expression level of miR-124 in Schwann cells was significantly increased,and the proliferation and migration of Schwann cells were promoted(P<0.01).Conclusion The expression level of miR-124 was increased gradually after sciatic nerve injury,may be involved in the process of nerve regeneration.
