骨髓基质细胞HS-分泌物白细胞介素-对人急性髓细胞性白血病细胞系HL-0抗凋亡的作用

摘    要:目的 探讨骨髓基质细胞HS-分泌物白细胞介素-6(IL-6)急性髓细胞性白血病(AML)细胞系HL-0增殖与凋亡的影响。方法 外培养HL-60细胞与HS-5细胞,构建共培养体系,应用扫描电子显微镜、ELISA法、细胞计数试剂盒8(CCK-8)法、AnnexinV-FITC/PI 双染流式细胞术、Real-time PCR和Western blotting技术分别检测HL-60细胞增殖和凋亡的变化;将不同组的HL-60细胞分别接种于BALB/c裸鼠,观察并记录成瘤情况。结果 骨髓基质细胞HS-5分泌的IL-6能够促进HL-60细胞增殖、抑制其凋亡,并下调HL-60细胞中的促凋亡基因Bax的表达,上调抗凋亡基因Bcl-2的表达。共培养组HL-60细胞在BALB/c裸鼠中成瘤能力最强,HL-60细胞单独培养组成瘤能力最弱。结论 骨髓基质细胞HS-5促进HL-60细胞增殖、抑制其凋亡的部分作用机制是通过分泌IL-6实现的。

Abstract:Objective To investigate the effect of interleukin( IL)-6 secreted by bone marrow stromal cells HS-5 on activity and apoptosis of human acute myeloid leukemia( AML) cells HL-60 and its possible mechanism. Methods HL-60 cells and HS-5 cells were cultured in vitro, and the co-culture system was established. Scanning electron microscope,ELISA,cell counting kit-8( CCK-8),Annexin V-FITC/PI,double-staining flow cytometry,Real-time PCR and Western blotting techniques were used to detect the changes of viability and apoptosis of HL-60 cells,respectively. HL-60 cells from different groups were inoculated into BALB/c nude mice to observe and record tumorigenesis. Results IL-6 secreted by bone marrow stromal cells HS-5 could enhance the viability and inhibit the apoptosis of HL-60 cells,and downregulate the expression of pro-apoptotic gene Bax and and up-regulate the expression of anti-apoptotic gene Bcl-2 in HL-60 cells. HL-60 cells in co-culture group had the strongest tumorigenicity in BALB/c nude mice,while HL-60 cells alone had the weakest tumorigenicity. Conclusion Part of the mechanism by which bone marrow stromal cells HS-5 promote the proliferation of HL-60 cells and inhibit their apoptosis may be through the secretion of IL-6.
