杜仲叶提取物对大鼠高脂性脂肪肝的实验观察

摘    要:目的:通过建立高脂血症脂肪肝动物模型,探讨杜仲叶醇提取物(alcohol extractive of Folium Eucommiae,AEFE)对脂肪肝的防治作用。方法:高脂饮食8周加维生素D36×10^5IU/kg分3d腹腔注射,建立大鼠高脂血症脂肪肝动物模型。各给药组从造模第3周开始分别灌胃给予洛伐他汀(4mg·kg^-1·d^-1)、AEFE(70、140和420mg·kg^-1·d^-1),共给药6周。各组每天灌服等量生理盐水,灌胃容积为0.5mL/100g体重。结果:AEFE用药6周后显示,各剂量组血清胆固醇、甘油三酯和游离脂肪酸、谷丙转氨酶和谷草转氨酶均较模型组明显降低(P〈0.05),肝组织丙二醛含量降低和超氧化物歧化酶活性升高,肝脂肪变性和炎症反应减轻,尤以中剂量组作用最明显(P〈0.01)。AEFE对肝内肿瘤坏死因子-α浓度无明显影响。结论:AEFE改善高脂性大鼠肝脏脂肪变性,此作用可能与其降血脂、抗氧化损伤有关。

Experimental Observation of Alcohol Extractive of Folium Eucommiae on the Rats with Hyperlipidemia Fatty Liver

Abstract:Objective.. To investigate the effects of alcohol extractive of Folium Eucommiae (AEFE) on the rats with hyperlipidemia fatty liver. Methods: Fatty liver model was established by feeding SD male rats with hyperlipid feed and vitamin D3 administration (6 ×10^5 IU/kg by intraperitoneal injected on 3 successive days) for 8 weeks. AEFE (70, 140 and 420 mg·kg^-1·d^-1,ig) was given for 6 weeks and was compared with lovastatin (4 mg·kg^-1·d^-1). Results: Compared with model group, AEFE not only decreased the elevations of levels in total cholesterols (mmol/L) (4.63±1.19 vs 2.54±0.58, 1.87±0.18 and 1.96±0.81), triglyceride (mmol/L) (3.16±0.57 vs 1.18±0.24, 0.98±0.17 and 0. 76±0.11), free fatty acids (mmol/L) (654.73±101.52 vs 505.38±89.64, 416.98±108.34 and 400. 854±93.56), alanine aminotrans-ferase (u/L) (56.32±6.11 vs 40.49±5.27, 31.43±5.14 and 29.17±4.51), aspartate aminotransferase (u/L) (62.54±5.79 vs 48.29±4.68, 32.04±3.15 and 32.33±4.04) in serum and malondialdehyde (nmol/mg pro) (7.31±0.79 vs 6.05±0.63, 4.98±0.72 and 5.72±0.82) in hepatic tissue significantly, but also reversed the decrease in superoxide dismutase activity (U/rag pro) (39.44±4.21 vs 47.03±6.16, 53.17±6.71 and 50.48±5.24) in hepatic tissue. At the same time, AEFE obviously ameliorated the pathologic changes on fat liver, but did not influence the levels of tumor necrosis factor-α in hepatic tissue. Conclusion: AEFE has a protective effect on hyperlipidemia fatty liver and its protection may involve in regulating plasma lipid metabolic disorder and enhancing anti-oxidant capacity.
