痹肿消汤对实验性关节炎大鼠血浆TNF-α及IL-1β的影响

摘    要:目的观察痹肿消汤(BZXD)对实验性关节炎大鼠血浆TNF-α和IL-1 β的影响,探讨痹肿消汤治疗类风湿性关节炎的作用机制.方法75只SD大鼠随机分为4组,皮下注射Ⅱ型胶原诱导实验性关节炎模型,采用放免法检测各组大鼠不同时间血浆TNF-α和IL-1 β水平.结果造模大鼠88%出现关节炎症状;造模25 d后,模型组、痹肿消汤组及甲氨喋呤组TNF-α和IL-1β水平明显高于正常组(P<0.05);且模型组的TNF-α和IL-1β水平明显高于痹肿消汤组及甲氨喋呤组(P<0.01);随着时间的延长,模型组TNF-α和IL-1 β水平逐渐升高,痹肿消汤组及甲氨喋呤组则逐渐降低;而痹肿消汤组TNF-α和IL-1 β水平低于甲氨喋呤组(P<0.05).结论TNF-α和IL-1 β在RA滑膜组织炎症形成和发展中发挥着重要作用;痹肿消汤能下调血浆TNF-α和IL-1 β的水平,其作用优于甲氨喋呤组.

Abstract:Objective: To observe the influence of BZXD on the plasma TNF-αand IL-1β in rats with C Ⅱ-induced arthritis (CIA) and explore the mechanism of BZXD in the treatment of rheumatoid arthritis . Methods: 75rats were randomly divided into four groups. The rat experiment arthritis model was established by subcutaneous injection with Ⅱ collagen and Freund' s complete adjuvant (FCA). The plasma TNF-αand IL-1β levels were detected with radio-immunity assay at different time. Results: The incidence of arthritis in rats immunized with C Ⅱ was approximately 88%. The plasma TNF-αand IL-1β levels of the model group, BZX decoction group and methotrexate (MTX) group were notably higher than those of the normal group (P <0.05). The plasma TNF-α and IL-1β levels of the model group were higher than those of the MTX control group and BZX decoction treatment group at different time (P <0.01). The plasma TNF-α and IL-1β levels rose step by step, but those of the BZX decoction group and MTX group decreased gradually. Moreover, the plasma TNF-αand IL-1 β levels of BZX decoction group were lower than those of MTX group (P <0.05). Conclusion: TNF-αand IL-1β play very important roles in the formation and development of RA disease. BZX decoction can notably decrease the plasma TNF-αand IL-1β levels, which is better than MTX.
