口腔鳞状细胞癌组织中基质金属蛋白酶-2与血管生成的关系

摘    要:目的 观察口腔鳞状细胞癌组织中基质金属蛋白酶 (matrixmetalloproteinase - 2 ,MMP - 2 )表达及其与微血管密度 (microvesseldensity ,MVD)的关系 ,探讨MMP - 2对口腔鳞状细胞癌血管生成的作用和意义。 方法 应用SABC免疫组织化学法检测 4 0例口腔鳞状细胞癌组织MMP - 2表达和MVD ,并分析MMP - 2表达与MVD及它们与口腔鳞状细胞癌组织临床病理特征之间的关系。结果 癌组织中MMP - 2高表达显著高于癌旁组织 ,癌旁组织MMP - 2低表达亦明显高于正常组织 (P <0 .0 5 )。MMP - 2高表达、MVD与口腔鳞状细胞癌病理分级和临床分期无关 ,与口腔鳞状细胞癌颈淋巴结转移密切相关 (P <0 .0 5 )。癌组织中MMP - 2高表达的MVD均显著高于MMP - 2低表达者 (P <0 .0 1)。结论 口腔鳞状细胞癌组织MMP - 2高表达与MVD及口腔鳞状细胞癌颈淋巴结转移有密切关系 ,MMP - 2可能是口腔鳞状细胞癌的重要促血管生成因子之一。

Abstract:Objective To detective the role and the significance of MMP-2 on angioenesis in oral squamous cell carcinoma(OSCC) and to observe the relationship between the expression of MMP-2 and microvessel density (MVD) in OSCC.Methods SABC immunohistochemical staining was used to detecte the expression of MMP-2 and MVD in various regions of OSCC and normal tissue.The relationship between the expression of MMP-2 and MVD as well as their relationship with clinicopathologic factors were analyzed.Results The expression of MMP-2 and MVD were negative in the normal tissue.The expression of MMP-2 in cancer tissue was significantly higher than that in the tissue adjacent to carcinoma(P< 0.05);The expression of MMP-2 in tissue adjacent to carcinoma was significantly higher than that in the normal tissue (P< 0.05).The high expression of MMP-2 in the OSCC tumor tissue was related to tumor invasion and neck lymph node metastasis.The MVD in the cases with high expression of MMP-2 was significantly higher than that in those with the low expression of MMP-2 in cancer tissue and tissue adjacent to carcinoma(P< 0.01).Conclusion The high expression of MMP-2 was significantly associated with MVD and neck lymph node metastasis of OSCC,MMP-2 might be one of the important factors in the angiogenesis of OSCC.
