摘    要:目的:建立怀牛膝药材的高效液相色谱(HPLC)指纹图谱。方法:色谱柱为SHIMADZU shim-pack VP-ODS(250mm×4.6mm,5μm),柱温为30℃,流动相为甲醇-0.1%醋酸水溶液(梯度洗脱),流速为1.0mL·min-1,检测波长为279nm。结果:建立了怀牛膝原药材和经硫磺熏制过的怀牛膝药材的指纹图谱。12批怀牛膝原药材有24个共有峰,其中9批药材的相似度都在0.9以上,另外3批的相似度较低;11批经硫磺熏制过的怀牛膝药材有20个共有峰,其中7批药材的相似度都在0.8以上,相似度较原药材低。结论:该方法简便、准确、专属性强,可用于怀牛膝药材的质量控制。

Abstract:OBJECTIVE: To establish an HPLC fingerprint of Achyranthes bientata. METHODS: SHIMADZU shim-pack VP-ODS C18 column(250 mm×4.6 mm, 5μm)was used with mobile phase consisted of acetonitrile-0.1% acetic acid (gradient elution) at flow rate of 1.0 mL·min^-1. The column temperature was 30 ℃ and the detection wavelength was set at 279 nm. RESULTS: HPLC fingerprints of A. bientata and sulphur-fumigated A. bientata were established. HPLC fingerprints of A. bientata showing 24 characteristic peaks were established from 12 batches ofA. bientata. The similarity of 9 batches was above 0.9 and the other 3 batches had low similarity. The HPLC fingerprints of 11 batches of A. bientata fumigated by sulphur showed 20 common characteristic peaks. The similarity of 7 batches of A. bientata fumigated by sulphur was above 0.8 and the similarity was lower than the original medicinal sample. CONCLUSION: The method is simple, accurate and specific, and it can be used for quality control of A. bientata.
