葛根醇提物对2型糖尿病模型大鼠的保护作用研究

目的:研究葛根醇提物对2型糖尿病模型大鼠的保护作用。方法:通过给予大鼠尾静脉注射链脲佐菌素,高脂饮食复制2型糖尿病大鼠模型。60只Wistar大鼠分为6组,即正常对照、模型、二甲双胍(70mg·kg-1)和葛根醇提物高、中、低剂量(150、100、50mg·kg-1)组。ig给药,每天1次,连续10周。通过测定空腹血糖(FBG)、血清胰岛素(Flns)含量,计算出胰岛素敏感性指数(ISI),测定肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)含量。结果:模型组ISI和TNF-α、IL-6含量显著高于正常对照组;葛根醇提物高、中剂量组ISI和TNF-α、IL-6含量显著低于模型组。结论:葛根醇提物可通过调节ISI和TNF-α、IL-6含量来改善2型糖尿病模型大鼠的症状,以为葛根醇提物的临床应用提供实验依据。

Protection Effect of Pueraria lobata Ethanol-extract on Type 2 Diabetes Model Rats

OBJECTIVE： To study the protection effect of Pueraria lobata ethanol-extract on type 2 diabetes model rats. METHODS： Rats were given intravenous injection of streptozotocin, high fat diet to establish rat model of type 2 diabetes mellitus. 60 Wistar rats were divided into 6 groups, ie normal control group, model group, metformin group （70 mg.kg 1）,p. lobata etha- nol-extract high-dose, medium-dose and low-dose （150, 100, 50 mg. kg1） groups. They were given medicine intragastrically once a day for 10 consecutive weeks. The insulin sensitivity index （ISI） was calculated by determining the contents of fasting blood glu- cose （FBG） and serum insulin （Flns）, and the contents of tumor necrosis factor （TNF-α） and interleukin 6 （IL-6） were deter- mined. RESULTS： The contents of ISI, TNF-α and IL-6 in model group were significantly higher than in normal control group; The contents of ISI, TNF-α and IL-6 in P. lobata ethanol-extract high-dose and medium-dose groups were significantly lower than in model group. CONCLUSION：P. lobata ethanol-extract can be obtained by adjusting the contents of ISI, TNF-α and IL-6 to im- prove the symptoms of type 2 diabetic model rats. The conclusions provide experimental basis for clinical use of P. lobata etha- nol-extract.
