| 摘 要： | 目的:建立测定北虫草发酵培养物中虫草素含量的方法。方法:采用反相高效液相色谱法。色谱柱为DIAMONSIL-C18(250mm×4.6mm,5μm),流动相为乙腈-水-甲醇(5∶90∶5),流速为0.8mL·min-1,柱温为40℃,检测波长为259nm。结果:虫草素检测浓度在2.56～51.20μg·mL-1范围内与峰面积积分值呈良好的线性关系(r=0.9998);平均回收率为98.35%,RSD=2.71%(n=6)。结论:本方法灵敏、快速、准确,适用于北虫草发酵培养物中虫草素的检测。

| Abstract: | OBJECTIVE： To establish the method for the content determination of cordycepin in fermented Cordyceps militaris. METHODS：RP-HPLC method was adopted.DIAMONSIL-C18（250 mm×4.6 mm, 5μm） column was used with acetonitrile-water-eth- anol （5：90：5） as mobile phase at the flow rate of 0.8 mL,min-1. The detection wavelength was set at 259 run and column temperature was 413 ℃ .RESULTS： The linear range of cordycepin was 2.56-51.20 μg· mL-1 （r=0.999 8） with an average recovery of 98.35% （RSD=2.71%, n=6）. CONCLUSION： The method is sensitive, rapid and accurate for the determination of cordycepin in fermented C. militaris. |
