丹参和川芎嗪联合应用对大鼠脑缺血再灌注氧化损伤的保护作用研究

摘    要:目的:研究丹参和川芎嗪联合应用对大鼠脑缺血再灌注氧化损伤的保护作用。方法:将Wistar大鼠200只随机分5组,即假手术、模型、丹参、川芎嗪、丹参+川芎嗪组。采用线栓法复制大鼠大脑中动脉缺血再灌注损伤模型,观察大鼠缺血再灌注后缺血脑梗死面积,脑水分、脑钙、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果:模型组脑梗死面积比假手术组显著增大(P<0.01);各用药组与模型组比脑梗死面积显著减小(P<0.01),且丹参+川芎嗪组效果更好;与假手术组比较,模型组MDA、脑钙和脑水分含量显著增高(P<0.01),而SOD活性显著降低(P<0.01);与模型组比较,各用药组脑钙和脑水分含量显著降低(P<0.05),SOD活性显著升高(P<0.05),且丹参+川芎嗪组效果更好。结论:丹参和川芎嗪联合应用比单用对大鼠脑缺血再灌注损伤后脑细胞保护作用更有效。

Study on the Protective Effect of Salvia miltiorrhiza Combined with Ligustrazine on Oxidative Damage in Rats with Cerebral Ischemia-reperfusion Injury

Abstract:OBJECTIVE: To investigate the protective effect of Salvia miltiorrhiza combined with ligustrazine on oxidative damage in rats with cerebral ischemia-reperfusion injury. METHODS: 200 Wistar rats were randomly divided into 5 groups: sham operation group, model group, S. miltiorrhiza group, ligustrazine group and combination of S. miltiorrhiza and ligustrazine group. The models of cerebral ischemia reperfusion injury model rats were established by the middle cerebral artery occlusion (MCAO). The changes of the cerebral ischemic area, the contents of brain wet, calcium, MDA, SOD activity in cerebral tissue were observed in rats after modeling. RESULTS: Compared with sham operation group, cerebral ischemic area of model group increased significantly (P〈0.01). Compared with model group, cerebral ischemic area of each medication groups decreased, there was significant difference (P〈0.01), especially in combination of S. miltiorrhiza and ligustrazine group. Compared with sham operation group, the contents of MDA, calcium and brain wet of model group increased significantly (P〈0.01) while SOD activity decreased (P〈0.01).Those parameters of each medication groups were significantly lower than model group (P〈0.05), SOD activities of each medication groups were higher than model group(P〈0.05), especially in combination of S. miltiorrhiza and ligustrazine group.CONCLUSION: The protective effect of S. miltiorrhiza combined with ligustrazine injection is better than them alone on cerebral ischemia reperfusion injury.
