半夏生物碱对人肝癌细胞Bel-7402增殖的影响

摘    要:目的:研究半夏生物碱对人肝癌细胞Bel-7402增殖的影响。方法:将Bel-7402与半夏生物碱(400、200、100μg·mL-1)共同培养,采用MTT比色法测定半夏生物碱对Bel-7402增殖的影响,TUNEL技术检测其对Bel-7402细胞凋亡的影响,免疫组化SABC染色法检测B细胞淋巴瘤/白血病-2(Bcl-2)和Bcl-2相关X蛋白(Bax)的表达情况。结果:作用24h后,半夏生物碱高、中、低剂量(400、200、100μg·mL-1)对Bel-7402增殖有显著抑制作用,且抑制作用随着药物浓度增加而加强,最高抑制率可达到36.98%。TUNEL结果显示,半夏生物碱高、中、低剂量(400、200、100μg·mL-1)能有效诱导Bel-7402细胞凋亡,且呈量效关系。半夏生物碱高、中、低剂量(400、200、100μg·mL-1)可使Bax表达显著升高而Bcl-2表达显著下降。结论:半夏生物碱对Bel-7402生长有一定的抑制作用,可诱导其凋亡,其作用机制可能为上调Bax和下调Bcl-2。

Abstract:OBJECTIVE: To study the effect of the alkaloids of Pinellia ternata on human hepatocarcinoma cell strain Bel-7402 proliferation. METHODS: Human hepatocarcinoma cell strain Bel-7402 co-cultured with alkaloids of P. ternata (400, 200, 100 μg. mL^-1). MTT method was used to detect the effect of alkaloids of P. ternate on human hepatocarcinoma cell strain Bel-7402 proliferation. TUNEL technique was used to detect the effect of alkaloids of P ternata on human hepatocarcinoma cell strain Bel-7402 apoptosis, and immunohistochemistry SABC staining method was used to test Bax and Bcl-2 protein expression. RESULTS: The high-dose, medium-dose and low-dose of alkaloids of P ternata (400, 200, 100μg · mL^-1) could inhibit the proliferation of Bel-7402 cell after 24 hours treatment, the inhibitory effect enhanced as the concentration increased, the highest inhibitory rate reached 36.98% in dose-response relationship. TUNEL showed that three different doses of alkaloids of P. ternata could induce Bel-7402 cell apoptosis in dose-response relationship. Three different doses of alkaloids of P. ternata could increase Bax expression but decrease Bcl-2 expression. CONCLUSION:Alkaloids of P. ternata can inhibit human hepatocarcinoma cell strain Bel-7402 proliferation and introduce its apoptosis, and its mechanism is associated with up-regulation of Bax and down-regulation of Bcl-2.
