HPV16、HPV18感染及p53蛋白过度表达与肺癌的相关性研究

摘    要:为研究人乳头瘤病毒16和18型(HPV16和HPV18)感染及p53基因突变与肺癌的关系,应用经病理学证实的肺癌活组织,行聚合酶链反应(PCR)检测癌组织中的HPV16、HPV18;并用LSAB免疫组化法检测相应蜡块标本中p53蛋白的过度表达。在48例肺癌组织标本中未检出HPV16、HPV18;p53蛋白表达呈阳性,阳性率为54.2%(26/48),其中鳞癌66.7%(18/27),腺癌57.1(4/7),小细胞癌33.3%(1/3),大细胞癌33.3%(2/6),类癌20.0%(1/5)。提示肺癌的发生可能与HPV16、HPV18感染无关,而与p53基因突变有关,尤以与鳞癌和腺癌关系密切

Abstract:The aim of this study was to gain some insights into the relationship between the human papillomavirus(HPV)type 16 and type 18 and the role that gene p53 products(p53)play in the carcinogenicity of HPVs.Every of our 48 human lung cancer cases was screened for the presence of HPV 16 and 18 DNA by polymerase chain reaction(PCR)and the expression of tumor suppressor gene p53 using immunohistochemistry assay in paraffin embedded formalin fixed sections.The HPV 16 and 18 DNA were not detected in these cases but p53 protein was detected in 26 cases using immunohistochemistry assays with the monoclonal antibody of mutant P53 protein,the total positive rate is 54.2%(26/48).In squamous lung carcinoma cases the positive rate is 66.7%(18/27)and in lung adenocarcinoma cases,57.1%(4/7).The p53 immunostain was positive in 1 out of 3 small cell lung cancers(33.3%),in 2 out of 6 large cell lung cancers(33.3%),in 1 out of 5 carcinocoid cases(20.0%),but was negative in all of the 10 control cases.It is indicated that tumor suppressor gene mutation has significant relationship with human lung cancer( P <0.05),p53 gene mutation plays an important role in the carcinogenicity of human lung cancer but is not related to the carcinogenicity of small cell lung cancer,large cell lung cancer and carcinocoid of lung cancer( P >0.05).
