褪黑素降低亚硒酸钠对小鼠的肝毒性

摘    要:目的研究褪黑素对亚硒酸钠致小鼠肝毒性的影响。方法 30只雄性昆明小鼠(体重18~20 g),随机分为3组,每组10只。对照组,每日灌胃生理盐水;亚硒酸钠组和亚硒酸钠+褪黑素组,每日灌胃亚硒酸钠6 mg/kg bw。2 h后,对照组和亚硒酸钠组灌胃2%的乙醇水溶液;亚硒酸钠+褪黑素组,灌胃褪黑素15 mg/kg bw(溶解在2%的乙醇水溶液)。连续6 d。检测小鼠血清转氨酶,肝脏丙二醛含量,总抗氧化能力,超氧化物歧化酶和过氧化氢酶活力、及肝脏谷胱甘肽过氧化物酶和谷胱甘肽硫转移酶活力,研究褪黑素对亚硒酸钠致肝毒性、氧化应激和调节含硒酶和二相酶的影响。结果与结论褪黑素可以减轻毒性剂量亚硒酸钠产生的氧化应激,降低亚硒酸钠的毒性,且不降低其预防癌症的疗效。

Abstract:Objective To investigate the protective role of melatonin against hepatic-toxicity induced by sodium selenite in mice.Method Thirty male Kunming mice(18-20 g) were randomly divided into three groups with 10 mice per group.The control group(CON) was administered water.The sodium selenite treated group(SS) and sodium selenite plus melatonin treated group(SS+MT) were administered sodium selenite at the dose of 6 mg/kg bw.After 2 h,the animals in CON and SS groups were administered water containing 2% ethanol,whiles SS+MT group was administered melatonin at the dose of 15 mg/kg bw(dissolved in water,containing 2% ethanol).All mice were treated by gavage for 6 d continuously.Transaminase activities in serum,level of malondialdehyde,total antioxidative competence,and the activities of superoxide dismutase,catalase,glutathione peroxidase and glutathione-S-transferase in liver were determined.Results and conclusion Melatonin confers resistance to oxidative stress induced by sodium selenite,and thereby to decrease toxicity of sodium selenite,without compromising its anti-tumor effect.
