基于网络药理学探讨丹参治疗骨质疏松症的作用机制

摘    要:目的:采用网络药理学方法分析中药丹参防治骨质疏松症(osteoporosis,OP)的药效物质基础及其分子作用机制。方法:通过TCMSP数据库筛选出丹参的活性成分及其作用靶点;检索OMIM、GeneCards和DisGeNET数据库获得OP的相关靶基因;绘制韦恩图获得丹参-OP共有靶点。通过Cytoscape软件构建疾病-药物-有效成分-靶点相互作用网络;利用Bioconductor工具对共有靶点进行GO功能注释和KEGG通路富集分析。结果:共获得OP相关基因2 026个,丹参抗OP潜在有效成分21个,包括丹参素、丹参酮ⅡA、隐丹参酮、木樨草素等,这些有效成分共作用于474个靶点,其中有53个与OP相关,其潜在调控信号通路139条,包括AGE-RAGE、PI3K-Akt、JAK-STAT等与各种骨细胞增殖、分化密切相关的信号通路。结论:丹参抗OP具有多成分、多靶点、多途径的作用特点,网络药理学为丹参抗OP作用机制的阐明提供了科学依据,并为中药的防病治病机制研究提供参考思路。

Abstract:Objective: To analyze the pharmacological substance basis and molecular mechanism of Chinese medicine Danshen( salvia miltiorrhiza) for the prevention and treatment of osteoporosis( OP) using network pharmacology. Methods: The active constituents of Danshen( salvia miltiorrhiza) and their action targets were screened out through the TCMSP database. The relevant target genes of OP were obtained by searching the OMIM,Gene Cards and Dis Ge NET databases. The Wein diagram was drawn to obtain the salvia miltiorrhiza-OP common targets. The disease-drug-active ingredient-target interaction network was constructed by Cytoscape software;the GO functional annotation and KEGG pathway enrichment analysis of common targets were performed using the Bioconductor tool. Results: A total of 2026 OP-related genes were obtained,and 21 potential anti-OP active ingredients of salvia miltiorrhiza were obtained,including tanshinol,tanshinone IIA,cryptotanshinone,luteolin,etc. These active ingredients acted on474 targets,of which 53 were related OP-related,its 139 potential regulatory signaling pathways,including AGE-RAGE,PI3 K-Akt,JAK-STAT and other signaling pathways closely related to the proliferation and differentiation of various bone cells. Conclusion:The anti-OP of Danshen( salvia miltiorrhiza) has the characteristics of multiple components,multiple targets,and multiple pathways. Network pharmacology provides a scientific basis for clarifying the anti-OP mechanism of Danshen( salvia miltiorrhiza) and provides reference ideas for the research on the mechanism of disease prevention and treatment of Chinese medicine.
