探究半夏不同炮制方法对半夏秫米汤治疗失眠的影响

摘    要:目的:基于半夏不同炮制方法探究半夏秫米汤治疗失眠的效果,筛选出最佳疗效品种及剂量,并探究其作用机制。方法:实验分2轮进行,均采用对氯苯丙氨酸腹腔注射法复制大鼠失眠模型,后予以中药干预。第1轮实验结束后通过翻正实验筛选出最优疗效组进行第2轮实验,第2轮实验结束后检测翻正实验,同时采用酶联免疫吸附(ELISA)法检测大鼠脑组织中5-羟色胺(5-HT)、去甲肾上腺素(NE)、白介素-1β(IL-1β)的浓度。结果:第1轮筛选实验结果显示,雌雄大鼠体质量阳性组和姜半夏组与模型组比较,差异均有统计学意义(P<0.01,P<0.05)。睡眠潜伏期缩短时间及睡眠持续时间方面,雌性大鼠阳性组、姜半夏组、生半夏组与模型组比较,差异均有统计学意义(P<0.01,P<0.05),其中阳性组>姜半夏组>生半夏组。雄性大鼠阳性组、姜半夏组与模型组比较,差异均有统计学意义(P<0.01,P<0.05),其中阳性组>姜半夏组。第2轮剂量实验结果显示,雌性大鼠体质量阳性组、姜高组、姜中组与模型组比较,差异均有统计学意义(P<0.01,P<0.05)。与模型组比较,阳性组、姜高组、姜中组可明显缩短大鼠睡眠潜伏期,延长睡眠持续时间,升高5-HT、IL-1β水平,降低NE水平,差异均有统计学意义(P<0.01,P<0.05),其中阳性组>姜高组>姜中组。结论:高剂量姜半夏组成的半夏秫米汤治疗失眠效果最优,其机制可能是通过调节下丘脑-垂体-肾上腺轴通路上神经递质的含量发挥作用。

Abstract:Objective:To investigate the effect of Banxia Shumi decoction in the treatment of insomnia based on the different processing methods of Pinellia ternate,to screen out the optimal processed product and dose,and to explore related mechanism of action.Methods:The experiment was carried out in two rounds.A rat model of insomnia was established by intraperitoneal injection of para-chlorophenylalanine,and then traditional Chinese medicine(TCM)intervention was given.After the first round ended,the turn-over test was used to screen out the group with the optimal outcome for the second round,and then the turn-over test was performed again after the second round ended.ELISA was used to measure the concentrations of 5-hydroxytryptamine(5-HT),norepinephrine(NE),and interleukin-1β(IL-1β)in brain tissue.Results:The results of the first round showed that there was a significant difference in the body weight of female and male rats between the positive group/the ginger-processed Pinellia ternate group and the model group(P<0.01 or P<0.05).There were significant differences in the shortening of sleep latency and the duration of sleep in female rats between the model group and the positive group,ginger-processed Pinellia ternate group,and unprocessed Pinellia ternate group(P<0.01 or P<0.05),among which the positive group had the best improvements,followed by the ginger-processed Pinellia ternate group and the unprocessed Pinellia ternate group;there were significant differences in these indices in male rats between the positive group/the ginger-processed Pinellia ternate group and the model group(P<0.01 or P<0.05),among which the positive group had better improvements than the ginger-processed Pinellia ternate group.The results of the second round showed that there was a significant difference in the body weight of female rats between the model group and the positive group,high-dose ginger-processed Pinellia ternate group,and middle-dose ginger-processed Pinellia ternate group(P<0.01 or P<0.05).Compared with the model group,the positive group and the high-and middle-dose ginger-processed Pinellia ternate groups had a significant reduction in sleep latency and a significant increase in sleep duration,as well as significant increases in 5-HT and IL-1βand a significant reduction in NE(P<0.01 or P<0.05),and the positive group had the best indices,followed by the high-and middle-dose ginger-processed Pinellia ternate groups.Conclusion:Banxia Shumi decoction with high-dose ginger-processed Pinellia ternate has the best clinical effect in the treatment of insomnia,possibly by regulating the content of neurotransmitters in the hypothalamic-pituitary-adrenocortical axis.
