参苓白术散对脾虚泄泻大鼠消化吸收功能的影响

摘    要:目的:探讨参苓白术散对脾虚泄泻大鼠消化吸收功能的影响。方法:选取雄性SPF级大鼠60只,采用生大黄水致泻结合饮食失节法复制脾虚泄泻模型,将造模成功的大鼠平均分为5组,即模型组、柳氮磺吡啶组(简称阳性组,0.5 g·kg^-1)、参苓白术散高(20 g·kg^-1)、中(10 g·kg^-1)、低(5 g·kg^-1)剂量组。连续给药14 d后,检测血浆中生长抑素(somatostatin,SS)、胃动素(motilin,MTL)、血管活性肠肽(vasoactive intestinal peptide,VIP)、血浆淀粉酶和尿液中D-木糖含量,并进行组间比较。结果:正常组大鼠皮毛富有光泽,精神状态良好;造模后大鼠明显消瘦,便溏,肛周污秽,毛发疏散、竖立,皮毛没有光泽。与正常组相比,造模后大鼠体质量明显下降,模型组大鼠血浆中MTL、SS、VIP、血浆淀粉酶活性指标和尿液D-木糖表达水平均明显下降,组间比较差异具有统计学意义(P<0.05)。给药后,阳性组及参苓白术散各给药剂量组均能不同程度地升高脾虚泄泻模型大鼠的体质量,其中参苓白术散高剂量组与正常组相比,差异无统计学意义(P>0.05);与模型组相比,各给药剂量组均能显著提高大鼠体质量、尿液D-木糖、血浆中MTL、SS、VIP、血浆淀粉酶活性,差异具有统计学意义(P<0.05)。与阳性组相比,参苓白术散高、中剂量组对大鼠体质量升高程度更为明显(P<0.05),参苓白术散低剂量组与阳性组作用强度相当,差异无统计学意义(P>0.05);参苓白术散高剂量组升高大鼠血浆MTL、SS、VIP强度与阳性组相当(P>0.05),参苓白术散中剂量组改善SS、VIP指标方面与阳性组比较,差异无统计学意义(P>0.05),而MTL指标则明显低于阳性组。与参苓白术散高剂量组相比,参苓白术散中、低剂量组均明显偏低。结论:参苓白术散能显著改善脾虚泄泻大鼠的消化吸收功能,其作用机制可能与上调脾胃激素MTL、SS、VIP表达水平,升高血浆淀粉酶活性有关。

Abstract:Objective:To investigate the effect of Shenling Baizhu Powder on digestive and absorption function in rats with diarrhea due to spleen deficiency.Methods:Sixty male SPF rats were selected to establish the diarrhea model of spleen deficiency by rhubarb water-induced diarrhea combined with diet disorder.The successful rats were divided into five groups on average:model group,sulfasalazine group(0.5 g·kg^-1),Shenling Baizhu powder high(20 g·kg^-1),medium(10 g·kg^-1)and low dose group(5 g·kg^-1).After 14 days of continuous administration,the contents of somatostatin(SS),motilin(MTL),vasoactive intestinal peptide(VIP),plasma amylase and D-xylose in urine were measured and compared between groups.Results:The normal group had glossy fur and good mental state.After modeling,the rats were obviously thin,loose stool,dirty around anus,and their hair was evacuated and erected,and their fur was not glossy.Compared with the normal group,the weight of rats decreased significantly after modeling.The levels of MTL,SS,VIP,amylase activity in plasma and D-xylose in urine decreased significantly in the model group(P<0.05).After administration,the weight of diarrhea model rats with spleen deficiency was increased in different degrees in the positive group and Shenling Baizhu powder dosage groups,and there was no significant difference between the highdose group and the normal group(P>0.05);compared with the model group,each dosage group could significantly increase the weight of rats,urine D-xylose,plasma MTL,MTL,MTL.SS,VIP and plasma amylase activity were significantly different between groups(P<0.05).Compared with the positive group,the weight gain of rats in the high and middle dose group was more obvious(P<0.05),and the effect intensity of the low dose group was similar to that of the positive group,but there was no significant difference(P>0.05);the effect of the high dose group was similar to that of the high dose group(P>0.05),and the effect of the medium dose group was different in improving the indexes of SS and VIP.There was no significant difference(P>0.05),but the MTL index was significantly lower than that of the positive group.Compared with the high dose group,the middle and low dose group was significantly lower.Conclusion:Shenling Baizhu Powder can significantly improve the digestive and absorption function of rats with diarrhea due to spleen deficiency.Its mechanism may be related to the up-regulation of MTL,SS,VIP expression and the increase of plasma amylase activity.
