淫羊藿对不同肾虚证模型小鼠肝脏功能的影响

摘    要:目的:探讨淫羊藿水煎液对不同肾虚证小鼠肝脏功能的影响。方法:将60只小鼠分为正常组,肾阳虚组和肾阴虚组,采用连续9 d肌肉注射氢化可的松25 mg·kg^-1·d^-1制备小鼠肾阳虚模型,隔天皮下注射氢化可的松50 mg·kg^-1·d^-1,4次制备肾阴虚动物模型,验模成功后,各组小鼠灌胃给予相同剂量的淫羊藿水提液。14 d后HE染色观察肝脏组织病理改变,化学试剂法检测血浆中天冬氨酸转氨酶(aspartate transaminase,AST)、丙氨酸转氨酶(alanine aminotransferase,ALT)、总蛋白(total protein,TP)、白蛋白(albumin,AL)、三酰甘油(triacylglycerol,TG)、总胆固醇(total cholesterol,TC)水平,RT-PCR法检测肝脏腺苷受体A1 mRNA表达。结果:与正常组比较,正常+淫羊藿组TP、TC显著升高(P<0.05)。与肾阳虚组比较,肾阳虚+淫羊藿组ALT显著降低(P<0.05),TP显著升高(P<0.05)。与肾阴虚组比较,肾阴虚+淫羊藿组TC显著升高(P<0.05)。给予淫羊藿后,各组小鼠肝脏腺苷受体A1 mRNA的表达有不同程度的下调,其中,肾阳虚组给予淫羊藿后下调的程度更明显。结论:淫羊藿对不同肾虚证模型小鼠肝脏功能的影响存在差异,可能与其减少肝脏腺苷受体A1mRNA的表达有一定关系。

Abstract:Objective:To investigate the effects of Epimedium on liver function in different kidney deficiency syndrome mice.Methods:60 mice were divided into normal group,kidney yang deficiency group and kidney yin deficiency group,and the model of kidney yang deficiency was prepared by intramuscular injection of hydrocortisone 25 mg·kg^-1·d^-1 for 9 consecutive days,the model of kidney yin deficiency were prepared by injecting hydrocortisone 50 mg·kg^-1·d^-1 4 times.After the model was successfully tested,mice of each group were given the same amount of Epimedium water extract for 14 days.The liver tissue pathology was observed by HE stain,chemical reagents to detect aspartate aminotransferase(AST),alanine aminotransferase(ALT),total protein(TP),albumin(AL),triglyceride(TG),total cholesterol(TC),RT-PCR was used to detect liver adenosine receptor A1 mRNA expression.Results:Compared with the normal group,TP and TC were significantly increased in the normal+Epimedium group(P<0.05).Compared with the kidney yang deficiency group,the kidney yang deficiency+Epimedium group′s ALT level was significantly decreased(P<0.05),and the TP was significantly increased(P<0.05).Compared with the kidney yin deficiency group,the kidney yin deficiency+Epimedium group′s TC level was significantly increased(P<0.05).After the administration of Epimedium,the expression of A1 R mRNA of each group was reduced,and the reduction of kidney-yang deficiency group was more obviously.Conclusion:Epimedium has different effects on the liver function of mice with different kidney deficiency syndrome,which maybe related to the reduction of hepatic adenosine receptor A1 mRNA expression.
