摘    要:目的:通过对蛴螬提取物干预实验性年龄相关性黄斑变性(age-related maculat degeneradon,AMD)实验研究,观察蛴螬提取物对实验眼脉络膜新生血管组织的趋化因子受体7(CXC chemokine receptor 7,CXCR7)及血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,同时比较口服、滴眼两种给药途径的差异。方法:建立病变易发生脉络膜新生血管(choroidal neovascularization,CNV)大鼠模型,将32只大鼠随机分成A、B、C、D 4组。A组:健康空白组(8只);B组:模型组(8只);C组:口服组(8只);D组:滴眼组(8只)。C组予以蛴螬提取物,按5.6 g·kg^-1,稀释至2 m L灌胃,每日1次;D组予以蛴螬滴眼液滴眼,每日4次;A、B、D组蒸馏水灌胃,每天1次,每次2 m L,A、B、C组予以玻璃酸钠滴眼液滴眼,每日4次。于造模后7 d、28 d行荧光素眼底血管造影检查,造模后28 d行免疫组化检测CXCR7与VEGF表达。结果:荧光素眼底血管造影检查显示:造模后第4周除B组眼底仍有荧光渗漏增多外,C、D组未见明显发展。CX-CR7与VEGF免疫组化结果显示:CXCR7与VEGF含量B组较A组显著上升,差异有统计学意义(P<0.05)。给药后,C、D组含量较B组明显减少,差异有统计学意义(P<0.05),C、D组含量接近,差异无统计学意义(P>0.05)。结论:蛴螬提取物可以抑制实验性CNV模型中CXCR7与VEGF的表达,从而可以抑制实验性CNV的形成。两种给药途径并无明显差异。

Abstract:Objective:To observe the effects of Qicao(Grub)extract on the treatment of experimental age-related maculat degeneradon(AMD),and to observe the CXC chemokine receptor 7(CXCR7)and the expression of vascular endothelial growth factor(VEGF).At the same time,the differences between the oral and eye drops were compared.Methods:A rat model of choroidal neovascularization(CNV)prone to lesions was established,and 32 rats were randomly divided into four groups:A,B,C,and D:Group A:Healthy blank group(8 animals);Group B:model group(8 animals);group C:oral group(8 animals);group D:eye drop group(8 animals).Group C was given Qicao(Grub)extract,diluted to 2 m L by gavage at 5.6 g·kg^-1,once a day;group D was given Qicao(Grub)drops,4 times a day;group A,B,and D distilled water 1 Each time,2 mL was administered to the stomach,and groups A,B,and C were given sodium hyaluronate eye drops 4 times a day.Fundamental fluorescein angiography was performed on the 7th and 28th days after modeling,and the expressions of CXCR7 and VEGF were detected by immunohistochemistry on the 28th day after modeling.Results:The fluorescein fundus angiography showed that in the 4th week after the model,except for the increase of the fluorescence leakage in the fundus of group B,there was no significant development in groups C and D.CXCR7and VEGF immunohistochemical results showed that the content of CXCR7 and VEGF in group B was significantly higher than that in group A,and the difference was statistically significant(P<0.05).After administration,the contents of groups C and D were significantly reduced compared with group B,and the difference was statistically significant(P<0.05).The contents of groups C and D were close,and the difference was not statistically significant(P>0.05).Conclusion:The Qicao(Grub)extract can inhibit the expression of CXCR7 and VEGF in the experimental CNV model,and thus can inhibit the formation of experimental CNV.There were no significant differences between the two routes of administration.
