薏苡仁酯对结肠癌SW480增殖和凋亡的影响

目的观察薏苡仁酯对结肠癌SW480增殖和凋亡的影响。方法结肠癌SW480细胞分为4组，其中A组为对照组，B，C，D组分别在培养基中给予5，10，20mg／L薏苡仁酯处理。采用噻唑蓝比色法检测4组细胞生长情况，流式细胞术检测细胞周期和凋亡情况。结果作用24h后，B，C，D组的抑制率分别为16．43％，22．51％，31．24％，呈剂量依赖关系，与A组比较差异均有统计学意义（P〈O．05）；给药后24hD组细胞G0～G，期比例较A组增高，S期细胞比例较A组降低（P〈O．05）；D组24，48h凋亡率明显高于A组，差异有统计学意义（P（0．05）。结论薏苡仁酯可在体外直接抑制结肠癌细胞的增殖，诱导凋亡，在结肠癌中药防治中有一定作用。

Effect of coixenolide on the proliferation and apoptosis of SW480 cells in patients with colon cancer

Objective To investigate the effect of coixenolideon on the proliferation and apoptosis of SW480 cells in patients with colon cancer. Methods The SW480 cells were divided into groups B, C, D （receiving 5 mg/L, 10 mg/L and 15 mg/L coixenolideon respectively） and group A（control group）. The effect of coixenolideon on the proliferation was observed with MTT technique, and the cycle and apoptosis of SW480 cells were examined with flow cytometry after 24 and 48 hours. Results The inhibiting rate was 16.43%, 9.2.51% and 31.24%respectively in groups B, C and D（P〈 0.05）, significantly different from that in group A （P 〈 0.05）. The inhibiting rate was dose dependent. After administration for 24 hours, the ratio of cell phase was higher in G0/G1 phase and lower in S-phase in group D than that in group A（P〈0.05）. The apoptotic rate was higher in group D than that in group A after 24 and 48 hours, which showed significant differences between two groups（P〈0.05）. Conclusion Coixenolideon inhibits the proliferation, induces apoptosis of SW480 cells, and plays a certain role in the prevention and treatment of colon cancer with traditional Chinese medicine.
