苦参碱对血小板衍生生长因子介导的大鼠肝星状细胞迁移、增殖和凋亡的影响

目的研究苦参碱对体外培养的肝星状细胞（HSC）迁移、增殖与凋亡的影响,探讨苦参碱抗肝纤维化的作用机制。方法用0.25、0.5mg/ml的苦参碱分别处理肝星状细胞-BB型（HSC-T6）和由血小板衍生生长因子-BB（PDGF-BB）干预的HSC-T6,观察HSC的生长情况,采用Transwell小室检测细胞迁移,用MTT法测定细胞增殖,TUNEL法检测细胞凋亡。结果苦参碱0.25mg/ml组和0.5mg/ml组细胞迁移数、吸光度值均低于空白对照组,细胞凋亡率则明显增高;在有PDGF-BB参与的3组中,苦参碱0.25mg/ml组和0.5mg/ml组的细胞迁移率和吸光度值降低,但是细胞凋亡率在3组之间差别无统计学意义。结论苦参碱对单纯激活状态下的HSC迁移、增殖均有较强的抑制作用,并能促进HSC凋亡的发生;苦参碱对PDGF作用下HSC的迁移、增殖有一定的抑制作用,但对此状态下的HSC的凋亡则无明显促进作用。

Effects of Matrine on Migration, Proliferation and Apoptosis of Rat Hepatic Stellate Cell Induced by PDGF in Vitro

Objective To investigate the effect of matrine on migration, proliferation and apoptosis of hepatic steilate cell （HSC） induced by PDGF in vitro. Methods HSC line was incubated separately with matrine in different concentration of 0.25 and 0.5 mg/ml, cell proliferation was assessed by MTT assay. Apoptosis was tested by TUNEL in situ assay. The migration ability of HSCs was observed with Transwell chamber assay. Results The cell migration rate and absorbance value of the groups with 0.25 mg/ml and 0.5 mg/ml matrine were significantly lower than those of the control group, as well as in PDGF groups. But there was no difference in rates of ceil apoptosis among three groups. Conclusion Matrine can inhibit HSC migration, proliferation and induce apoptosis of HSC, which might be one of the mechanisms that matrine counteracts against liver fibrosis. But there was no effect of matrine on HSC apoptosis induced by PDGF.
