| 摘 要： | 探讨ＨＳＶ１－ＴＫ基因在人肾癌ＧＲＣ－１细胞中表达的可能性及意义。方法采用基因工程技术构建带ＨＳＶ１－ＴＫ基因的逆转录病毒重组体Ｇ１Ｎ－ＴＫ、用脂质体法对人肾癌ＧＲＣ－１细胞进行转染及用新霉素筛选。结果成功克隆出ＨＳＶ１－ＴＫ基因的ＧＲＣ－１／ＴＫ细胞，并对无环鸟苷敏感。

| Abstract: | Objective To investigate the expression and significance of HSV1 TK gene in human renal cancer GRC 1 cells . Methods A retrovirus expression vector G1Na TK was constructed with pgk tk cDNA and a retroviral vector G1Na.The G1Na TK vector was transfer into renal cancer GRC 1 cells with lipofectin reagent and the HSV TK vector producer cell line GRC 1/TK was isolated in 1mg/ml neomycin medium. Results The HSV TK vector producer cell line GRC 1/TK was obtained which confered a chemosensitivity to ACV. Conclusions HSV1 TK gene might be used in gene therapy of human renal cancer GRC 1 cells. |
