冷水束缚应激下脾虚大鼠胃黏膜H~+,K~+-ATP酶活性及mRNA的表达变化

摘    要:目的观察脾虚大鼠模型在冷水束缚应激负荷下,胃黏膜损伤程度与H+,K+-ATP酶活性及基因表达的变化。方法 SD大鼠随机分为正常组、脾虚组、应激组、脾虚应激组。采用100%大黄水煎液灌胃10 d造成脾虚模型。冷水束缚应激负荷6 h后观察各组大鼠胃黏膜损伤指数,用无机磷法测定胃黏膜H+,K+-ATP酶的活性;用RT-PCR法检测胃黏膜H+,K+-ATP酶mRNA的表达。结果脾虚大鼠胃黏膜肉眼未见明显损伤,应激及脾虚应激组大鼠胃黏膜均出现点状及条索状损伤,且脾虚应激组损伤程度更为显著。脾虚组H+,K+-ATP酶活性及mRNA的表达率较正常组低(P均<0.05);应激及脾虚应激组H+,K+-ATP酶活性及mRNA的表达较正常组高(P均<0.05);且脾虚应激组大鼠H+,K+-ATP酶活性及mRNA的表达增高幅度有所增大。结论脾虚大鼠在冷水束缚应激状态下胃黏膜易损伤性增高,其机制之一可能与脾虚应激后胃黏膜H+,K+-ATP酶功能升高有关。

Abstract:Objective It is to observe the injury degree of gastric mucosa,and the changes of enzyme activity and the mRNA expression of the H+,K+-ATP ase in gastric mucosa of spleen deficiency rats under stress in cold water.Methods SD rats were randomly divided into normal group,spleen deficiency group,stress group and spleen deficiency stress group.The spleen deficiency group and the spleen deficiency stress group were given 100% rhubarb decocted by oral for 10 days to establish spleen deficiency model.The stress group and the spleen deficiency stress group were put in cold water for 6 hours,and then the gastric mucosa ulcer index was measured by Guth method,the activity of H+,K+-ATP ase in gastric mucosa by the inorganic phosphorus method and the mRNA expression of H+,K+-ATP ase by RT-PCR method.Results No obvious injury of gastric mucosa was found under naked eyes in spleen deficiency group,while point injury and streak injury were found in the stress group and the spleen deficiency stress group,and the injury was more obvious in the latter group.Compared with the normal group,the activity and the mRNA expression of H+,K+-ATP ase in stress group and the spleen deficiency stress group were higher(P<0.05) while the spleen deficiency group were lower(P<0.05),and the increasing degree of the activity and the mRNA expression of H+,K+-ATP ase in the spleen deficiency stress group were higher significantly than those in the stress group.Conclusion The occurrence of the more vulnerability of the gastric mucosa in the spleen deficiency rats may be related to the rising of gastric mucosa H+,K+-ATP ase in spleen deficiency rats after stressing.
