摘    要:目的探讨脂多糖(LPS)促进磨屑诱导分泌溶骨性因子 M-CSF、IL-1在人工关节松动中的作用.方法采集30例人体外周血,分离单核细胞(MOs),分组培养,A组(对照组):仅 MOs;B组:MOs+超高分子聚乙烯微粒(UHMWPE,未去除LPS);C组:MOs+LPS;D组:MOs+UHMWPE (去除LPS);E组:MOs+UHMWPE(去除LPS)+LPS.培养48h后,放免法测定上清中巨噬细胞集落刺激因子(M-CSF)、白介素1(IL-1)的含量.结果 E组 M-CSF、IL-1含量明显高于A、B、C、D组(P<0.05);B组M-CSF、IL-2较D组增高(P<0.05).结论 LPS具有促进UHMWPE刺激M-CSF、IL-1分泌的作用,增强了UHMWPE的溶骨效应.

Effect of LPS on osteolysis induced by wear particles in vitro

Abstract:Objective To explore the possibility of Lipopolysaccharide(LPS) contribute to aseptic loosening of orthopedic implants. Methods Peripheral blood samples were obtained from healthy human donors.Monocytes(MOs) were isolated and assigned to 5 groups.Group A was treated with MOs alone,group B with MOs + ultrahigh molecular weight polylethylene(UHMWPE) debris,group C with MOs + LPS(100ng/ ml),group D with MOs + UHMWPE(LPS removal),group E with MOs + U HMWPE(LPS removal) + LPS(100ng/ ml).The cells were co-cultured for 48 hours and the mixtures were tested for osteolytic factors includingM-CSF IL-1. Results The levels of these factors were much higher in group E than in the other 4 groups.There was significant difference between group B and group D(P<0.05). Conclusions LPS can stimulate MOs to secrete osteoclast-activating cytokines,LPS and UHMWPE cooperative interact to enhance the biological effects of aseptic loosening.
