健脾理气法对功能性消化不良大鼠的治疗作用及其机制的研究

摘    要:目的]观察健脾理气法对脾虚气滞型功能性消化不良(FD)大鼠的治疗作用及其分子生物学机制。方法]60只SPF级Wistar雄性大鼠随机分为正常组,治疗组,对照组,模型组。后3组采用夹尾刺激法制备FD模型,各组分别以0.85%氯化钠、健脾理气中药、0.85%氯化钠、安慰剂灌胃,1周后检测大鼠胃动力,胃窦及血清中钙调蛋白激酶Ⅱ(CaMKⅡ)的含量以及胃窦细胞外信号调节激酶1/2(ERK1/2)磷酸化水平。结果]与正常组相比,模型组大鼠胃动力降低,血清及胃窦中CaMKⅡ含量降低,胃窦ERK1/2磷酸化水平降低;经健脾理气中药治疗后,大鼠胃动力明显改善,血清及胃窦中CaMKⅡ含量有所恢复。结论]CaMKⅡ的表达及ERK1/2的活性与FD存在一定的关系,二者含量和活性的降低可能是FD发病的机制之一;健脾理气法不影响ERK1/2的活性,但可能通过增加CaMKⅡ表达,促进胃动力,达到治疗FD目的。

Abstract:Objective]To investigate the effects of Jianpi Liqi Decoction on rats with functional dyspepsia(FD) and explore its underlying mechanism.Methods]Sixty SPF Wistar male rats were randomly divided into four groups:normal group,Jianpi Liqi Decoction intervention group,control group and model group.Except the normal group,the other groups were established FD models by stimulating rat tail.Rats were given saline,Jianpi Lipi Decoction,saline and placebo respectively.After a week of administration,gastric dynamics,the level of CaMKⅡ in gastric antral muscle and serum and the phosphorylation level of ERK1/2 in gastric antral muscle were detected.Results]Compared to normal group,gastric dynamics,the level of CaMKⅡ in gastric antral muscle and serum,the phosphorylation level of ERK1/2 in gastric antral muscle were decreased in model group,while Jianpi Liqi Decoction improved gastric dynamics and reversed the decrease of CaMKⅡ in gastric antral muscle and serum.Conclusion]The level of CaMKⅡ and the activity of ERK1/2 are related to FD,whose decrease may be one of the underlying mechanisms of this disease.Jianpi Liqi Decoction does not regulate ERK1/2 activity,but plays its therapeutic roles for FD by increasing the level of CaMKⅡ to promote gastric dynamics
