糖尿病大鼠血管性痴呆模型的建立

摘    要:目的:建立合适的慢性实验性糖尿病大鼠血管性痴呆(VaD)模型。方法:腹腔注射链脲佐菌素(STZ)诱导慢性实验性糖尿病(DM)大鼠模型,永久性结扎双侧颈总动脉制作VaD模型。行为学筛选合格的SD大鼠分为假手术组(Sham)、DM组、VaD组和DM VaD组。观察术后2周、4周、8周大鼠的一般状况,并应用Y-迷宫观察其空间定向学习和记忆的能力。结果:DM组手术4周后出现学习记忆能力下降。VaD组和DM VaD组在术后2周即出现学习记忆能力下降,并呈逐渐下降趋势;DM VaD组较同期的VaD组下降更为严重。结论:STZ诱导的糖尿病大鼠经颈动脉结扎后成功建立了VaD模型,可用以进一步研究糖尿病与VaD发病的关系。

Abstract:Aim: To establish a suitable animal model of vascular dementia(VaD) with experimental diabetes mellitus(DM).Methods: Diabetes mellitus was induced in male SD rats by an intraperitoneal injection of streptozotocin. The bilateral common carotid arteries were ligated permanently to establish rat models of VaD. The rats reaching standard were randomly divided into four groups: sham control group,sham DM group, VaD group,and DM+VaD group.The weight and serum glucose were recorded. The spacial study and memory in Y-maze after 2,4,8 weeks of operation were estimated. Results: The rats in VaD group and DM+VaD group showed worse performance 2 weeks after operation,and 4 weeks after operation in DM group. The rats in DM+VaD group had more error numbers (EN) and longer total reaction time (TRT) than VaD group at the same time. Conclusion: The VaD rat model with DM has been established successfully, which can be used to study the relation between diabetes and VaD.
