电损毁致大鼠痉挛性脑性瘫痪动物模型制备及其鉴定

摘    要:目的:探讨不同条件下建立电毁损锥体束致大鼠一侧肢体痉挛性脑性瘫痪(CP)模型方法,并对CP模型动物进行鉴定。方法:将60只大鼠随机平均分为对照组A、实验组B和C。均沿颅顶矢状缝作纵行切口,暴露前囟,选择前囟后6.6mm,矢状缝左侧1.0mm钻孔。对照组A不通电;实验组B通直流电2mA,20s/次,通电1次;实验组C通直流电2.5mA,30s/次,通电2次;电毁损锥体束制备模型,术后对大鼠行神经行为学检测(随意运动、姿势、肌张力等),并对鉴定为CP的大鼠观察1个月。结果:实验组C模型成功率(16/20例)及痉挛性瘫痪症状的稳定性明显高于实验组B(7/20例),差异有显著性(P〈0.05);CP大鼠存在明显随意运动障碍及姿势、肌张力异常。结论:直流电(恒流2.5mA,30s/次,通电2次)毁损锥体束可成功制备大鼠痉挛性CP动物模型,CP鼠具有神经行为学异常。

Abstract:Objective To establish one-side extremity spastic cerebral palsy model in rats by electronic injury of pyramidal tract and identify the animal model,and to identify with spastic cerebral palsy model rats.Methods Sixty Wistar rats were divided into three groups(A:normal control group;B and C:experiment group,n=20 in each group).The rats' pyramidal tract in groups B were injured by electric current 2 mA for 20 s,once,but rats in group C were injured by electric current 2.5 mA for 30 s,twice.The neural behaviors of all animals were tes-ted.Rats with cerebral palsy were checked for 30 days.Results Sixteen rats in group C(16/20 cases)developed cerebral palsy,but only 7 rats developed cerebral palsy in group B(7/20 cases),no rat in control group developed cerebral palsy.Cerebral plasy rats showed abnormality of posture and muscle tension.Conclusions An animal model for investigation of spastic cerebral palsy is established by electronic injury of pyramidal tract.A method for identifying rats with spastic cerebral palsy is successfully established.
