摘    要:目的探讨丙戊酸钠(VPA)对体外培养的人肝癌细胞系SMMC-7721细胞增殖抑制作用及对乙酰肝素酶(HPA)和增殖细胞核抗原(PCNA)表达的影响及机制。方法肝癌细胞株SMMC-7721分对照组和实验组。对照组细胞常规培养,实验组细胞施加VPA干预,设浓度梯度和时间梯度。四甲基偶氮唑蓝(MTT)比色法测定细胞增殖活性;RT-PCR半定量法测定细胞HPA及PCNA mRNA表达的变化;Western-blot法测定细胞HPA及PCNA蛋白含量。结果 VPA对SMMC-7721细胞的生长具有明显的增殖抑制作用,且表现为时间依赖性和浓度依赖性(P<0.05);当药物浓度增加至≥2.0mmol/L时,其HPA和PCNA mRNA及蛋白的表达量较对照组明显下调,且呈时间依赖性(P<0.05)。结论 VPA对肝癌SMMC-7721细胞有明显的增殖抑制作用,且能够在核酸和蛋白水平上下调HPA及PCNA的表达,从而抑制肿瘤的增殖、侵袭和转移。

Effect of sodium valproate on the proliferation of human hepatic cancer cell line-SMMC-7721 and expression of HPA and PCNA

Abstract:Objective To investigate the inhibitory action of sodium valproate(VPA) on the proliferation of human hepatic cancer cell line-SMMC-7721 and expression of HPA and PCNA,and to explore its action mechanism.Methods The SMMC-7721 was divided into control group and trial group.The cells in control group were cultured by routine method,however,the cells in trial group were cultured with VPA intervention,with concentration gradients and time gradients.The cell proliferation was determined by MTT assay,the expression levels of HPA and PCNA were detected by RT-PCR and the protein contents of HPA and PCNA in the cells were measured by Western blot.Results The sodium valproate inhibited obviously the proliferation of SMMC-7721 cells in a dose-and time-dependent manner(P<0.05).The expression levels of HPA and PCNA mRNA and protein were down-regulated in a time-dependent manner in SMMC-7721 cells when the VPA concentration was higher than or equal to 2.0mmol/L(P<0.05).Conclusion VPA has obvious inhibitory action on the proliferation of SMMC-7721 cells,furthermore,which can down-regulate the expressions of HPA and PCNA mRNA and protein,as a result,which can inhibit the proliferation,invasion and metastasis of tumors.
