呼吸道生物被膜菌致肺气虚证大鼠模型的研制

摘    要:目的:研制大鼠肺气虚证模型,为研究因慢性呼吸道感染导致肺气虚证的诊治提供可靠的实验动物。方法:健康SD大鼠随机分为空白对照组、烟熏对照组,以及高、中、低菌量模型组。模型组采用呼吸道生物被膜菌(bacterialbiofilm,BF)铜绿假单胞菌滴鼻法,辅以寒冷疲劳刺激引起大鼠呼吸道慢性感染,诱导肺气虚证的发生。观察各组证候表现、游泳时间、体重变化、血液流变学指标的改变,以及肺支气管组织器官病理改变。结果:高、中、低菌量模型组大鼠的一般体征均符合肺气虚证,症状较烟熏组稳定。游泳时间和体重增加低于空白对照组,尤以高菌量组为明显(P〈0.05,P〈0.01)。血流变指标提示血液流变性有障碍,肺及支气管炎症明显,炎症的程度和感染菌量有关。结论:将呼吸道生物被膜菌(铜绿假单胞菌)以滴鼻法并辅以寒冷疲劳刺激,可以诱导大鼠肺气虚证模型,相关的指标表明证候表现与中医学理论的病机相符,且该模型具有稳固、易复制、贴近原型的特点。
