摘    要:目的:建立检测大鼠牙周组织IL-6mRNA表达的原位杂交方法,弄清咬合力与牙周组织IL-6mRNA表达变化的关系,初探其在牙周组织改建中的分子机理。方法:选用大鼠为实验对象建立动物模型,采用地高辛标记的寡核苷酸探针的原杂交方法,检测不同咬合力状态下,大鼠牙周组织IL-6mRNA表达的变化。结果:牙周组织成纤维细胞和成骨细胞中均有IL-6mRNA阳性信号,而在阴性对照、空白对照中均未检测到信号。结论:本实验所建立的原位杂交方法具较高特异性的灵敏性。此方法用于口腔修复生物力学研究中心,从分子水平阐明了咬合力对牙周组织改建影响的机理,揭示了牙周组织结构与功能的关系。

Abstract:Objective The aim of this study is to establish method of in situ hybridization for the expression of IL -6mRNA in rat periodotium.To clarify the relationship berween bite force and IL- 6mRNA expression in periodontium, and elucidate the molecular mechanism of periodontium remodeling. Method Wista rats were selected to establish animal model by extracting the left maxillary molars. Oligonucleotide probe labeled with digoxigenin were used in in situ hybrid - ization (ISH) technique. ISH was applied to study the influence of bite force on the IL- 6mRNA expression in the rat pe riodontium. Results IL- 6mRNA positive reaction was observed in periodontal ligament fibroblasts and alveolar bone os-teoblasts. There were no IL - 6mRNA expression wsa observed in negative control experiment. Conclusion In situ hybridization method established in this study showed showed specificity and sensitivity. Application of ISH to study the oral prosthetics biomechanics, it helped to eludicate the molecular mechanism of the influence of bite force on periodontium remodeling, and also to better manifest the relationship between periodontium struture and mechanical condition.
