杂合肽CAMEm诱导鼻咽癌细胞凋亡的作用

摘    要:目的研究杂合肽(cecropinA-melittinhybridpeptidemutant,CAMEm)对鼻咽肿瘤细胞的诱导调亡作用。方法选取鼻咽癌CNE2细胞株为靶细胞,3T3细胞为正常细胞,以MTT比色法和Hoechest33342染色法,应用流式细胞仪和DNA凝胶电泳观察CAMEm对CNE2细胞的诱导调亡作用,检测杂合肽对CNE2细胞和3T3细胞生长抑制作用的时效和量效关系。结果与对照组相比,当5mg/LCAMEm作用12h时,抑制率升高,P<0·05;125mg/LCAMEm作用12h时,抑制率显著升高,P<0·01。0·5mg/L的CAMEm作用12h时,可以观察到调亡小体的产生;0·5、5、10和25mg/LCAMEm诱导CNE2细胞的调亡率分别达14·60%、17·42%、21·80%和36·10%,诱导调亡作用与剂量和时间呈明显正相关。CAMEm对3T3细胞没有显著影响。结论杂合肽CAMEm对CNE2细胞株具有明显诱导调亡作用,而且对正常细胞没有显著杀伤作用。

Abstract:OBJECTIVE:To investigate the anti-tumor effects of cecropin A-melittin hybrid peptide mutant in human nasopharyngeal carcinoma cell line in vitro. METHODS: The selected human nasopharyngeal carcinoma cell line CNE2 and 3T3 cell were treated with CAMEm at various concentrations and for different time. Growth suppression and apoptosis related to alterations were evaluated by MTT method,Hoechest33342, flow cytometry (FCM) and agarose gel electrophoresis. RESULTS: Campared with the control group, the vability repression rate of the treatment group with 5 mg/L CAMEm for 12 hours CAMEm was higher P<0.05 by MTT; the viability repression rate of the treatment group with 5 mg/L CAMEm for 12 hours CAMEm was significant higher, P<0.01. After the treatment with 0.5 mg/L CAMEm for 12 hours,apoptosis with nuclear chromatin condensation and fragmentation as well as cell shrinkage and the formation of apoptotic bodies were observed by using fluorescent microscope. For CNE2 cells treated with 0.5,5,10 and 25 mg/L CAMEm, the apoptosis rates were 14.60%,17.42%,21.80% and 36.10% in a dose-and-time-dependent manner. CAMEm had no effect on human 3T3 cell. CONCLUSION: CAMEm induces apoptosis of human nasopharyngeal carcinoma cell line CNE2, but shows little toxicity against human 3T3 cell.
