| 摘 要： | 目的通过基因检测和组织染色的方法,来研究丹参酮IIA磺酸钠对兔耳增生性瘢痕中MMP-1mRNA表达的影响和其对增生性瘢痕的作用。方法①先建立兔耳增生性瘢痕模型,以曲安奈德为阳性对照,分组分次瘢痕基底局部给药,取下瘢痕组织,使用RT-PCR方法检测细胞内金属基质蛋白酶MMP-1基因表达;用苏木素-伊红染色（HE）、苦味酸酸性复红染色法（VG）染色法对成纤维细胞和胶原纤维进行观察。结果 RT-PCR基因检测显示丹参酮及曲安奈德组的MMP-1mRNA的表达显著高于空白对照组;HE染色显示空白对照组中成纤维细胞数量明显高于丹参酮IIA磺酸钠组,丹参酮IIA磺酸钠组与曲安奈德组无明显差异;VG染色显示丹参酮IIA磺酸钠组胶原纤维排列较空白对照组显著整齐,与曲安奈德组无明显差异。结论丹参酮IIA磺酸钠能够通过增强MMP-1mRNA的表达及抑制成纤维细胞的增殖,减少胶原纤维的合成而抑制瘢痕的过度增生。
