摘    要:目的探讨张应力加载下骨髓基质细胞力生长因子(MGF)表达的影响及量效关系。方法骨髓基质细胞原代培养后加力不同时间经形态学观察,ELISA检测各组BMSC培养上清中ALP的浓度,Western blot检测加力后骨髓基质细胞MGF的表达。结果机械拉伸刺激下BMSC的形态学特性逐渐发生改变。加载0h、1h、3h、6h、12h、24h位移为1mm,频率为0.5Hz的机械张应力,均可增强BMSC的ALP活性,其中24h时ALP OD值最高(P〈0.05)。Western Blot分析BMSC加力后MGF蛋白的表达,结果为细胞加力后MGF在1h开始有变化,MGF表达水平随加力作用时间延长而增高,组间比较差异有统计学意义(P〈0.05)。结论 MGF可能通过刺激骨髓基质细胞ALP的分泌而促进其向成骨方向分化。
