库仑电化学法快速测定单胺类神经递质的方法学研究

摘    要:目的建立库仑阵列电化学高效液相色谱法快速测定大鼠脑组织中的单胺类神经递质的含量。方法采用库仑阵列电化学检测器检测去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA),采用Phenomenex C18反相色谱柱(150mm×4.60mm,5μm),流动相为缓冲液-乙腈(体积比87∶13),流速为1mL.min^-1,柱温为35℃。结果 4种单胺类神经递质的最低检测限为0.01515~0.01815ng,回收率为85.3%~100.5%。结论该法具有快速、准确、灵敏、样品制备简便等优点,可用于大鼠脑组织中单胺类神经递质的分离检测。

Abstract:Objective To establish a rapid method for determining monoamine neurotransmitters in rats' brain tissue by HPLC with coularray detector.Methods HPLC was performed on a Phenomenex C18 column(150×4.60 mm,5 micron). The mobile phase,consisted of acetonitrile-water phase(13:87),was at a flow-rate of 1 mL·min^-1. The column temperature was kept at 35 ℃. The levels of NE,DA,5-HT and 5-HIAA were determined.Results The minimum measurement limit was 0.015 15-0.018 15 ng. The average recoveries were 85.3 %-100.5%.Conclusion This method is rapid,accurate,highly sensitive and easy to operate. The procedure is suitable for determining the monoamine neurotransmitters in rats' brain tissue.
