二十味沉香丸预防低氧性肺动脉高压时对蛋白激酶C通路的影响

摘    要:探讨二十味沉香丸预防慢性低氧造成的肺动脉高压时对蛋白激酶C的影响及意义。方法:Wistar大鼠30只,随机分为3组(n=10):①正常对照组;②高原低氧组;③药物干预组,接受不同的干预,共30天。测各组大鼠肺动脉压,小动脉平滑肌细胞核密度,肺组织PKC活性检测及蛋白的表达。结果:药物干预组与正常对照组比较,肺动脉压高(P<0.05),但与高原低氧组相比较,肺动脉显著降低(P<0.05)。药物干预组、高原低氧组小动脉平滑肌细胞核密度显著高于正常对照组(P<0.05)。高原低氧组、药物干预组鼠肺组织中的PKC活性较正常对照组显著性增高(P<0.05)。高原低氧组、药物干预组较正常对照组PKC的表达显著性增高(P<0.05)。结论:二十味沉香丸可能通过增强蛋白激酶c通路的表达缓解慢性低氧造成的肺动脉高压形成。

Abstract:To investigate the preventive effects of PKC in pulmonary arterial hypertension caused by chronic hypoxia after treated by Tibetan twenty Chen Xiang Compound pill. Method :30 Wister rats were divided into three groups; normal group, hypoxia group and treatment group. Pulmonary artery pressure, SMC density of Pulmonary artery, activity and expression of PKC in lung tissue were detected by Western blot and immunohistochemistry, respectively. Result: Pulmonary artery pressure in treatment group was higher than that of normal group (P〈0.05 ), but lower in hypoxia control group ( P〈0.05 ). SMC density and PKC expression in hypoxia group and treatment group were significantly higher than that of normal group ( P 〈0.05 ). Conclusion: Tibetan twenty Chen Xiang Compound pill may prevent hypoxia pulmonary arterial hypertension by increasing expression of PKC pathway.
