摘    要:目的探讨植物雌激素木质素类物质去甲二氢愈创木二酸(NDGA)对乳腺癌细胞系MCF-7中雌激素受体(ER)α、ERβmRNA的调节作用,及对其增殖的影响,探讨木脂素类植物雌激素是否通过选择性ER调节剂的机制起作用。方法培养乳腺癌细胞系MCF-7,用不同浓度NDGA处理细胞系3 d后,采用实时荧光定量逆转录聚合酶链反应(RT-PCR)方法,观察不同剂量NDGA对两种ER亚型表达的影响。结果NDGA中等、较大剂量时对MCF-7细胞系的增殖有明显抑制作用。实时荧光定量PCR结果表明NDGA处理细胞系3 d后ERα表达先下降后上升,ERβ表达渐上升。结论木脂素类植物雌激素NDGA明显抑制乳腺癌细胞系MCF-7的增殖,且能够改变其ER不同亚型的表达,ER表达趋势改变提示NDGA具有ER调节剂的作用。

Abstract:Objective: To study the effect of nordihydroguaiaretic acid(NDGA) on the proliferation of MCF-7 cell line and its regulation on the expression of ERα and ERβ mRNA.Methods: Human breast cancer cell line MCF-7 was treated with different concentrations of NDGA(10,20,50,100 μmol/L) for 3 days.The proliferative status was assayed by MTT and the mRNA levels determined by real-time quantitative PCR.Results: NDGA displayed an inhibitory effect on the growth of MCF-7 cells.NDGA may increase the levels of ERβ mRNA in a dose-dependent manner.The levels of ERα mRNA in MCF-7 cells could be increased by lower concentrations of NDGA,but decreased by higher concentrations of NDGA.Conclusions: The results suggest that NDGA might serve as a potential selective estrogen receptor modulator(SERMs) for the prevention and treatment of cancer.
