摘    要:目的:研究单纯高甘油三酯血清(HTGBS)对人脐静脉内皮细胞(HUVEC)的损伤作用。方法:体外培养HUVEC,观察不同浓度的HTGBS对血管内皮细胞活力(MTT法测吸光度A值)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)活性,丙二醛(MDA)和一氧化氮(NO)含量的影响。运用WesternBlot和酶联免疫吸附测定法(Enzyme-Linked Immunosorbent Assay,ELISA)检测各组细胞中血红素氧化酶-1(HO-1)蛋白的表达和细胞培养上清液中碳氧血红蛋白(COHb)的含量。结果:5%浓度的HTGBS对血管内皮细胞有明显的损伤作用,显著降低细胞活力、NO含量和SOD、NOS活性(P<0.01),显著升高血管内皮细胞中LDH、iNOS活性和MDA含量(P<0.01),并且5%HTGBS显著诱导细胞HO-1蛋白的表达并增加细胞培养液中CO的生成量(P<0.01)。结论:HTGBS体外诱导HUVEC氧化损伤,使细胞活力和抗氧化能力显著降低,增加其通透性及脂质过氧化程度,影响内皮细胞分泌功能并能诱导HO-1蛋白表达代偿性升高。

The Injury of the Human Umbilical Vein Endothelial Cells Caused by High Triglyceride Blood Serum

Abstract:Objectives: To investigate the injuries caused by pure high triglycerides blood serum (HTGBS) on human umbilical vein endothelial cells(HUVECs ). Methods: Culture the HUVECs in vitro and observe the activities of HUVECs (the value of Absorbance)with different concentrations of HTGBS using MTT method. The activities of LDH, SOD and NOS as well as the contents of MDA and NO in the cells were detected. The expressions of HO-1 protein were detected using Western Blot. The relative amount of CO released into the media was quantitated as COHb by ELISA. Results: HTGBS of 5% in concentration could injure obviously the HUVECs, decrease the activity of SOD and NOS and the content of NO (P 〈0.01 ), and increase significantly the LDH and iNOS activities in endothelial cells and the content of MDA (P 〈0.01 ). The expression of HO-I protein in HUVECs and the content of CO in the media were increased significantly by HTG.Conelusion: HTGBS of high concentration could directly injure the structure and partial function of HUVECs, decrease the cell activity and antioxidant capacity, increase the permeability and lipid peroxidation, affect the metabolism of HUVEC and induce the componsatory increase in the expression of HO-lprotein.
