摘    要:目的 通过在内侧视前区(mPOA)微量注射NMDA,观察其对异丙酚睡眠作用的影响,探讨异丙酚睡眠作用机制。方法 SD大鼠42只,随机分为6组:NS组、2%二甲基亚砜(DMS0)组、异丙酚组、NMDA 10pmol组、NMDA 20pmol组、异丙酚+NMDA 20pmol组,记录并分析睡眠EEG变化。结果 微量注射异丙酚80 ng显著增加2 h内总睡眠时间,主要为延长非快动跟睡眠时期,缩短睡眠潜伏期。给予不同剂量NMDA时,2 h内总睡眠时间显著缩短,睡眠潜伏期延长。NMDA20pmol可拮抗异丙酚诱发的睡眠,主要缩短非快动眼睡眠时期,对快动眼睡眠无明显影响。结论 在mPOA微量注射NMDA可拮抗异丙酚的睡眠作用。

Abstract:Objective: To investigate the effects of NMDA on sleep inducing by propofol (Prop) mi-croinjection into the medial preoptic area (mPOA) and to explore the mechanism. Methods: 42 Sprague-Dawley (SD) rats were divided randomly into 6 groups: NS group, 2% DMSO group, prop, group, NMDA 10 pmol group, NMDA 20 pmol group, Prop + NMDA 20 pmol group. Subsequent electroen-cephalographic (EEG) sleep behavior was observed. Results: Microinjection of propofol 80ng significantly increased total sleep time (TST) and decreased sleep latency during 2 h after injection. The NREM sleep was primarily increased. Both doses of NMDA significantly decreased TST and increased sleep latency in 2 h. NMDA 20 pmol exerted its effect primarily by reducing NREM sleep episodes induced by propofol and not by reducing the REM sleep. Conclusions: The NMDA receptor in the mPOA is an important mediator for regulation of sleep-wakefulness and propofol-induced sleep.
