附子对腺嘌呤造成的小鼠慢性肾衰的疗效及其机制探讨

摘    要:目的:研究中药附子对腺嘌呤造成的小鼠慢性肾衰的疗效,并探讨其作用机制。方法:选取雄性小鼠随机分为4组:对照组(C),模型组(M),附子低剂量组(FL),附子高剂量组(FH)。对照组给予溶剂,模型组灌胃给予腺嘌呤,各给药组在给予腺嘌呤的基础上,再分别给予3g/kg/d、10g/kg/d的附子水煎剂。实验结束时,测定血清尿素氮、肌酐、γ-谷氨酰转肽酶,肾组织γ-谷氨酰转肽酶、一氧化氮、谷胱甘肽、自由基等的含量,并记录附睾精子数、全血血红蛋白含量。结果:附子高剂量组优于低剂量组,能降低尿素氮、肾系数,提高精子数、血红蛋白含量。而附子低剂量组则无明显疗效。结论:附子高剂量显示对腺嘌呤造成的慢性肾衰有一定的疗效,其作用机制还有待于进一步研究。

Abstract:Objective: To observe the effect and mechanism of Fuzi on chronic renal failure in mice.Methods: Male mice were randomly divided into four groups,that is,normal control group,model group(adenine,250 mg/kg every two days in the first week,130mg/kg every two days in the second and third weeks),Fuzi high dose group(10g/kg/d),Fuzi low dose group(3g/kg/d).At the end of the experiment,we measured the content of blood urea nitrogen(BUN),creatine(Cr),γ-glutamyltranspeptidase(γ-GT) in blood serum.The kidney were homogenated for measuring γ-GT,nitric oxide(NO) and glutathione(GSH).Besides,we recorded the number of sperm in the left epididymis and the content of hemoglobin in blood.Results: the high dose Fuzi could reduce the blood urea nitrogen,renal factor,increase sperm count,hemoglobin content,while the low dose Fuzi could not.Conclusions: Fuzi high dose group showed some efficacy,the mechanism needs to be studied further.
