摘    要:目的:探讨碱性成纤维细胞生长因子(bFGF)、脑源性神经生长因子(BDNF)对大鼠脑缺血损伤后海马区(SGZ)神经干细胞原住增殖的影响。方法:将Wistar大鼠84只随机分为空白对照组(12只)、bFGF组(24只)、BDNF组(24只)、bFGF+BD—NF组(24只)。采用线辁法制作局灶性大脑中动脉闭塞(MCAO)模型。分别于缺血再灌注后3d、7d、14d、28d处死大鼠。免疫组织化学方法动态检测SGZ区BrdU、Nestin的表达。结果:各组SGZ区的BrdU和Nestin阳性细胞均在脑缺血3d后开始增加,7d达到高峰,14d后开始下降,28d降至最低水平;与空白对照组相比,均有统计学意义(P〈0.05);其中bFGF+BDNF组Brdu和Nestin阳性细胞数增加更明显。结论:侧脑室注射bFGF、BDNF可促进脑缺血大鼠SGZ区内源性神经干细胞原位增殖;bFGF和EGF联合应用对脑缺血大鼠神经干细胞原位增殖效应有协同作用。

Effects of Bfgf and EGF on neural stem cell situ proliferation of MCAO rat hippocampus

Abstract:Objective:To investigate effects of basic fibroblast growth factor (bFGF) and brain--derived neurotrophic factor (EGF) on the neural stem cell situ proliferation of MCAO rat hippocampus after cerebral ischemic injury. Methods: A total of 48 adult wistar rats were selected and randomly assigned into a normal control group with 12, a bFGF group, a BDNF group, a bFGF and BDNF group with 24 in each group. Rat models of focal middle cerebral artery occlusion were established by the thread method in each group. Rats were killed at 3, 7, 14 and 28 days following ischemia/reperfusion. Immunohistochemistry was used to detect BrdU and Nestin expression in the rat hippocampus region. Results:Nestin and BrdU -- positive cells began to increase in the rat hippocampus region on Day 3 and reached a peak on Day 7, but decreased on Day 14 and reduced to a minimal levels on Day 28 after ischemia/reperfusion in each group. The number of positive cells was significantly increased compared with normal control group (P 〈0. 05). The number of Nestin and BrdU -- positive cells of the bFGF and BDNF group was significantly increased, more than that of the bFGF group and the BD-NF group. Conclusion :BDNF and bFGF injected into the lateral cerebral ventricle can activate endogenous neu- ral stem cell situ proliferation in the cerebral ischemic rat hippocampus region, and bFGF and BDNF have a synergistic effect in it.
