γ-hGH对乳腺癌细胞MCF-7的放射敏感性研究

摘    要:目的探讨重组人生长激素(γ-hGH)对乳腺癌细胞株MCF-7的放射敏感性。方法本实验分对照组、照射组、γ-hGH作用组及联合作用组,利用流式细胞仪测定细胞凋亡率,活细胞计数测定细胞存活率,利用克隆形成法测定细胞抑制率。结果流式细胞仪检测显示,照射组、作用组及联合作用组的凋亡率分别为:1.06%±0.48%、0.96%±0.38%,10.8%±3.46%,联合作用组高于任何1个单因素处理组(P〈0.05);照射组、作用组及联合作用组的细胞存活率分别为51.7%±1.10%、57.7%±1.20%和11.0%±1.00%,联合作用组下降最为显著(P〈0.01),细胞克隆形成抑制率分别为20.9%±0.65%、26.0%±1.10%和67.9%±0.93%,联合作用组高于任何1个单因素处理组(P〈0.01)。结论γ-hGH可以有效提高射线对乳腺癌细胞的放射敏感性。

The Radiative Sensitivity of the Breast Cancer Cell Treated by Recombinant Human Growth Hormone

Abstract:Objective To investigateon the radiosensitivity of breast cancer cell line MCF-7 treated by recombinant human growth hormone(γ-hGH).Methods The study contained control group,irradiation group,γ-hGH treatment group and combined therapy group.Apoptosis was analyzed by flow cytometry.Cell viability was dertemined by Trypan blue exclusion.Inhibition rate was calculated by colony formation test.Results The apoptosis rates in the irradiation group,γ-hGH treatment group and combined therapy group were 1.06%±0.48%,0.96%±0.38% and 10.8%±3.46%,respectively.The apoptosis rate in the combined therapy group was significantly higher than that in the other two groups(P0.05).The cell survival rates in the irradiation group,γ-hGH treatment group and combined therapy group were 51.7%±1.1%,57.7%±1.2% and 11.0%±1.00%,And cells in the combined therapy group had significantly lower survival rates than that in the other two groups(P0.05).Inhibited rates in the three groups were 20.9%±0.65%,26.0%±1.10% and 67.9%±0.93%,respectively,and the combined therapy group had significantly higher inhibited rates than the other two groups(P0.01).Conclusion γ-hGH could enhance the radiosensitivity of human breast cancer cell line MCF-7.
