特发性胎儿生长受限患者胎盘组织中松弛素受体的表达

摘    要:目的探讨特发性胎儿生长受限(FGR)患者胎盘组织中松弛素受体LGR7表达与胎儿生长受限发生的关系。方法应用链霉菌抗生物素蛋白.过氧化物酶连接法和RT—PCR技术检测20例正常孕妇(正常组)及20例特发性胎儿生长受限组(FGR组)胎盘组织中松弛素受体LGR7及mRNA的表达.结果①两组孕妇的胎盘组织中均有松弛素受体LGR7表达,其主要定位于胎盘绒毛滋养细胞的细胞膜上,在细胞滋养细胞、合体滋养细胞中都有较高水平的表达。②正常组胎盘组织中松弛素受体LGR7蛋白的表达水平为0.928±0.002,FGR组为0.656±0.024,两组比较,差异有统计学意义(P〈0.01)。③正常组织胎盘组织中松弛素受体LGR7 mRNA表达水平为0.780±0.021,FGR组为0.412±0.062,两组比较,差异有统计学意义(P〈0.01)。结论FGR患者胎盘纽织中松弛紊受体表达水平下降,可能与FGR的发病有关。

Expression of Relaxin Receptor in Placental Tissues of Pregnancy with Idiopathic Fetal Growth Restriction

Abstract:Objective To study the location and expression level of relaxin receptor in placenta tissues of preeclampsia and normal pregnancy, and investigate the relationship between relaxin receptor with idiopathic fetal growth restriction ( FGR). Methods Twenty placenta tissue samples from pregnant women with FGR ( study group ) , and 20 samples from normal pregnancies ( control group) were obtained. The expression level of relaxin receptor was detected by immunohistochemistry and the relaxin receptor mRNA by RT-PCR. Results The leucine-rich repeat-containing G protein-coupled receptor( LGR7 ) was positively expressed in both of the two groups . Relaxin receptor was located in the membrane of trophoblast cells. Strong signals was found in both of cytotrophoblastic cells and syneytiotrophoblast. The expression level of relaxin receptor in the control group was 0.928 ~ 0.002, and O. 656 :i: 0.024 in FGR group. The difference between the two groups was significant( P 〈0.01 ). In the control group, the level of relaxin receptor mRNA was 0. 780 ± 0. 021 ;in FGR group, it was 0.412 ± 0. 062. The difference was significant( P 〈 0.01). Conclusion The decrease in the expression level of relaxin receptor in placenta is related with the occurrence and development of FGR.
