CIK细胞联合NaHCO3抑制人肝癌裸鼠移植瘤生长的研究

摘    要:目的 将NaHCO3口服与CIK细胞(cytokine-induced killer,CIK)联合应用以提高CIK细胞在裸鼠体内的抗肝癌活性.方法 将HePG2-Luc细胞皮下移植入裸鼠皮下构建荷瘤小鼠模型,再分四组进行实验:(1)CIK输注+普通水喂养;(2)CIK输注+ 碱性水喂养;(3)生理盐水输注+碱性水喂养;(4)生理盐水输注+普通水喂养.随后定期用小鼠活体成像仪观察肿瘤组织生长速度及荧光强度,30d后取肿瘤组织称重,并制作病理切片,免疫组化检测肿瘤组织CD3及穿孔素的表达情况.结果 CIK输注联合碱性水喂养组的肿瘤荧光强度、肿瘤生长速度都明显低于其他各组,联合组肿瘤组织的CD3阳性及穿孔素阳性细胞数也多于其他组,差异有统计学意义(P<0.05),而碳酸氢钠单独处理组与对照组差异无统计学意义(P>0.05).结论 NaHCO3碱性水能有效增强CIK细胞抗肝癌的活性.

Abstract:Objective To improve the anti-tumor activity of CIK cell against hepatocellular carcinoma HePG2 cells,the bearing tumor nude mice were treated with NaHCO3 oral administration and CIK cell injection.Methods Nude mice were xeno-grafted with HePG2-luc cells and divided into four groups which were CIK cell injection plus NaHCO3 feeding,CIK cell injection plus drinking water feeding,normal saline injection plus NaHCO3 feeding ,and normal saline injection plus drinking water feeding.Then the tumor growth speed and fluorescence intensity were observed with IVIS Spectrum System.At 30 days,the tumor tissues were took out and the weights were detected.Finally,the tumor tissues were made into pathological section and the expression of CD3 and Perforin were detected with immunohistochemical.Results Both the growth speeds and fluorescence intensity of HePG2-luc cells were remarkably depressed when CIK cells combined with NaHCO3 compared with other groups.NaHCO3 feeding augmented the infiltration of CD3+ and Perforin+ T lymphocytes into the tumor mass.However,there were no statistical significantly differences between NaHCO3 feeding alone and control group.Conclusion NaHCO3 alkaline water can effectively enhance the anti-tumor activity of CIK cells against hepatocellular carcinoma HePG2 cells.
