摘    要:目的 探索使用新的纯物理加工工艺制备脾脏生物多肽,并通过调节免疫力药效学实验比较其与已上市产品的差异,为优化工艺、提高产品得率、生产出高效低毒产品提供参考依据.方法 采用低温循环微波、超声波、高速匀质机结合侧向膜超滤/过滤技术等物理方法制备一定分子量大小范围的脾脏生物多肽.建立环磷酰胺诱导小鼠免疫力低下模型,分灌胃和皮下注射两种途径给予小鼠自制脾脏生物多肽,测定小鼠抗体生成细胞水平、脾脏和胸腺脏器系数、血清细胞因子TNF-α和IL-2的表达水平,与阳性对照药脾氨肽对比评价脾脏生物多肽调节免疫力的效果.结果 本实验所制得的脾脏生物多肽,相对分子质量约为7 500,通过不同给药途径给药,与模型组相比,小鼠的抗体生成细胞、TNF-α及IL-2表达水平均比模型组显著增加(P<0.05),其中,灌胃组的TNF-α,皮下注射组的TNF-α、IL-2均优于阳性对照组.结论 成功探索出一种新的纯物理方法制备脾脏生物多肽的技术工艺,所得产品能显著拮抗环磷酰胺所致的免疫力低下,改善免疫功能,为新产品的开发奠定了基础.

Abstract:Objective To develop a new physical method for preparation of spleen polypeptides with increased production and quality.Methods Spleen polypeptides were prepared by a series of physical methods including low-temperature cycle microwave,ultrasonic wave,high speed mixing and filtration.Immunocompromised mice were established by subcutaneous injection of cyclophosphamide,and treated with spleen polypeptides by intragastric administration and subcutaneous injection.The antibody-producing cells,organ coefficients and serum cytokines were examined.Results The molecular weight of spleen polypeptides was about 7 500Da.Compared with the model group,the number of antibody-producing cells and levels of TNF-α and IL-2 were significantly increased in spleen polypeptide-treated mice (P<0.05).Conclusion Spleen polypeptides are successfully produced by a pure physical method and display immunoregulatory function in immunocompromised mice.
