葛根素抑制人子宫颈癌细胞株HeLa细胞增殖研究

目的 探讨葛根素(puerarin)对人子宫颈癌细胞株HeLa的增殖抑制和凋亡诱导作用,并探讨其机制.方法 MTT法检测不同浓度葛根素(0,12.5,25,50)处理48 h对HeLa细胞增殖的影响;流式细胞术检测葛根素对HeLa细胞凋亡的影响.Western blotting 检测葛根素对HeLa细胞Wnt,β-catenin ,Bax,Bcl-2,p53和p21表达水平影响.结果 葛根素对 HeLa细胞增殖有明显的抑制作用,呈明显的剂量-效应依赖性;葛根素明显促进HeLa细胞凋亡;Western blotting检测显示,经葛根素处理72 h后,Wnt,β-catenin ,Bcl-2,p53和p21表达明显下降,呈浓度依赖性.结论 葛根素可诱导HeLa细胞增殖抑制和凋亡, 其作用机制与抑制Wnt/β-catenin 信号通路相关.

Study on Puerarin inhibiting the proliferation of Human cervical cancer cell lines HeLa cells

Objective To explore the effect of Puerarin on proliferation and apoptosis of human Cervical Cancer Cells lines HeLa,and explore the potential mechanism.Methods After the HeLa cell was treated with Puerarin (0,12.5,25,50) for 48h,MTT method was used to evaluate the proliferation and the percentage of apoptotic cells were determined by flow cytometric analysis.The expression of Wnt,β-catenin,Bax,Bcl-2,p53 and p21 were detected by western blotting (WB).Results The results showed that puerarin treatment can significantly induce the proliferation inhibition and apoptosis of HeLa cell in concentration-dependent manner.Furthermore,that puerarin can decrease the expression of Wnt,β-catenin,Bcl-2,p53 and p21 in HeLa cell by WB methods.Conclusion Puerarin can induce apoptosis and proliferation inhibition of human Cervical Cancer Cells lines HeLa and the mechanism is associated with suppressing Wnt/β-catenin signalling.
