微流控芯片三相层流用于人参中人参皂苷的提取研究

摘    要:目的 建立基于三相层流微流控芯片萃取技术进行人参药材中成分提取的方法.方法 设计了三通道入口萃取芯片,两侧通道经表面疏水处理,用注射泵引入脂溶性溶剂乙醚及萃取剂水饱和正丁醇,中间亲水通道用注射泵引入药物初提液;将水饱和正丁醇所得提取液用高效液相色谱法(HPLC)进行测定,检测人参总皂苷的质量分数.结果 三相层流芯片比双相层流芯片所得人参皂苷质量分数高,3种人参皂苷Rg1、Re、Rb1的质量分数分别提高了3.95倍、3.32倍、5.94倍.结论三相层流芯片可用于3种不同极性溶剂的同时层流处理,效率高、时间短、消耗低,不但可用于同时提取和分离,也可推广至多种极性部位的共同萃取.

Abstract:Objective To establish a method for extraction of components from panax ginseng based on three-phase laminar microfluidic chip extraction technique. Methods A three-channel inlet extraction chip was designed. The two sides of the channel surface was treated by selective hydrophobic modification, the fat-soluble solvent and the extraction solvent were introduced into the chip by using the syringe pumps. The initial drug solution was introduced into the middle hydrophilic channel by using the syringe pump. After the extraction of the three-phase laminar chip, the content was determined by high performance liquid chromatography ( HPLC) . Results Compared with the two-phase laminar chip extraction, the three-phase laminar chip extraction yielded a higher ginsenoside content. The contents of three kinds of ginsenosides Rg1 , Re and Rb1 were increased by 3.95 times, 3.32 times, 5.94 times. Conclusion The three-phase laminar flow chip could be used for the simultaneous laminar flow of three different polar solvents with high efficiency, short time and low consumption, which not only could be used for simultaneous extraction and separation, but also to promote a variety of polar parts of the common extraction.
