金银花提取物对LPS致ARDS大鼠肺炎症影响的研究

目的 探讨金银花提取物对脂多糖(LPS)诱导的急性呼吸窘迫综合征(ARDS)模型大鼠的抗炎作用及其机制.方法 以LPS诱发的ARDS大鼠作为急性炎症动物模型,实验分为空白组、模型组、金银花组和激素组,观察各组肺组织的形态以及炎症细胞和炎症因子的变化,研究金银花的抗炎作用.结果 金银花提取物能改善ARDS大鼠肺组织病理损伤情况,抑制由炎症刺激物质LPS诱导的IL-1、IL-6和TNF-α炎症介质和细胞因子的释放.结论 金银花提取物对ARDS大鼠具有显著的保护作用,其机制可能与其降低肺泡灌洗液中促炎细胞因子IL-1、IL-6和TNF-α的释放和减轻组织脏器的损伤有关.

Effects of Lonicera japonica extract on lung inflammation in LPS-induced ARDS rats

Objective To investigate the effects of Lonicera japonica extract on lung inflammation in LPS-induced ARDS rats and elucidate the anti-inflammatory mechanisms of Lonicera japonica extract.Methods The ARDS model was established by LPS stimulation in SD rats.Forty healthy SD rats were randomly divided into 4 groups:the normal group,the ARDS model group,the Lonicera japonica extract group and the dexamethasone treatment group.The pathologic changes of lung tissue were observed in each group.Total cell numbers in broncho alveolar lavage fluid (BALF) were counted,and the concentrations of IL-1,IL-6 and TNF-α in BALF were measured by ELISA. Results Lonicera japonica extract improved sepsis-induced lung tissue lesion.Compared with the ARDS group,total inflammatory cell number in BALF and the level of IL-1,IL-6 and TNF-αin BALF in the group treated with Lonicera japonica extract were significantly decreased. Conclusion The results of this study revealed that Lonicera japonica extract could attenuate sepsis through suppression of IL-1,IL-6 and TNF-αinflammatory factors.Therefore,our findings suggest that Lonicera japonica extract might be useful as a potential therapeutic agent for treatment of sepsis.
