罗汉果水提物对非酒精性脂肪肝炎小鼠肠道菌群影响分析

摘    要:目的 探讨罗汉果水提物干预非酒精性脂肪肝炎(NASH)小鼠模型肠道菌群的变化.方法 C57BL/6小鼠随机分为蛋氨酸胆碱正常含量饲料(MCS)组、蛋氨酸胆碱缺乏饲料(MCD)模型组和MCD饲料联合罗汉果水提物处理的低、中、高浓度组[50、100、500 mg/(kg·d)],每组各10只.采用MCD饲料和等热量的MCS对照饲料喂养12 d后,采集粪便,送样进行16SrDNA V4V5 区测序分析.解剖小鼠,取肝脏组织进行HE染色检测脂肪变性及炎症.结果 小鼠肝脏病理切片HE染色观察发现MCD可诱导小鼠肝脏明显脂肪变性和炎性浸润,而罗汉果水提物干预可减少肝脏脂质和炎性浸润的现象.罗汉果水提物干预组中拟杆菌门对厚壁菌门的比值与MCD模型组的比值相比有较为明显的增高作用,且罗汉果水提物浓度越高越接近于对照组的比值.与模型组相比,罗汉果水提物处理组疣微菌门的Akkermansia muciniphilia菌属显著增加,高浓度罗汉果水提物组Akkermansia muciniphilia菌属高于对照组.结论 罗汉果水提物可以明显改善NASH小鼠的肠道菌群.

Abstract:Objective To investigate the changes of gut microboitas in the NASH mouse model treated by water extract of Siraitia grosvenorii.Methods C57BL/6 mice were randomly divided into the following five groups and received the corresponding treatment for 12 days: (1) mice fed with MCS diet;(2) mice fed with MCD diet;(3)-(5) mice fed with MCD diet supplemented with 50 ,100,500 mg/(kg·d) water extract of Siraitia grosvenorii,respectively.Each group has 10 mice.MCD diet and equal calorie MCS control diet for 12 days,the fecal samples were collected and sequenced for 16S rDNA V4V5 region.Liver tissue was subjected to HE staining to detect steatosis and inflammation.Results The analysis of pathology showed that the liver tissues of water extract of Siraitia grosvenorii treatment groups were minor lipid accumulation and inflammatory infiltration compared with that of the NASH model group.Compared with the MCD group,the ratio of Bacteroides and Firmicutes in Siraitia treatment groups significantly increased.And the Akkermansia muciniphilia of the warts microbacteria was significantly higher than that of the control group.Conclusion Siritia can significantly regulate the intestinal microflora in NASH mice.
